摘要
目的探讨匹诺塞林脑保护的作用机制。方法采用SH-SY5Y细胞制备OGD/R损伤模型,缺糖缺氧2 h、复糖复氧12 h,采用MTT法检测细胞活力及LDH释放,细胞核Hoechest33324染色检测细胞凋亡,免疫荧光检测细胞内GRP78、CHOP、P-eIF2α及ATF4表达的变化。结果匹诺塞林能提升OGD/R损伤的SH-SY5Y细胞的活力,减少LDH释放,提高GRP78表达,降低CHOP、ATF4及P-eIF2α的表达。结论匹诺塞林能减轻缺糖缺氧/复糖复氧诱导的SH-SY5Y细胞损伤,可能与其调节ERS凋亡途径的PERK-eIF2α-ATF4通路相关蛋白表达有关。
Objective To explore the mechanism of pinocerin's brain protection.Methods SH-SY5Y cells were used to establish OGD/R injury model with glucose deprivation and hypoxia for 2 hours and glucose reoxygenation for 12 hours,and LDH release were detected by MTT assay.Cell apoptosis was detected by Hoechst 33324 staining.The expression of GRP78,CHOP/GADD153,ATF4,and P-eIF2αin SH-SY5Y cells was detected by immunofluorescence.Results Pinocembrin can increase the cell viability while decreasing the release of LDH,and increase the expression of GRP78 but decrease the expression of CHOP/GADD153,ATF4,and P-eIF2αin SH-SY5Y cells.Conclusion Pinocembrin can alleviate SH-SY5Y cell injury induced by oxygen-glucose deprivation/reperfusion,and its mechanism may be related to the regulation of proteins expression on perk-eIF2α-ATF4 pathway about ERS apoptosis.
