摘要
沉默信息调节因子2相关酶1(SIRT1)是依赖于烟酰胺腺嘌呤二核苷酸的第Ⅲ类组蛋白去乙酰化酶,通过对靶蛋白的去乙酰化作用发挥生理学效应。在心血管系统中,SIRT1参与调节线粒体功能、氧化应激、炎症反应、细胞凋亡与自噬等过程。SIRT1的活性异常与心力衰竭、心肌缺血再灌注损伤、心肌肥厚、糖尿病心肌病、动脉粥样硬化等心血管疾病的发生与发展密切相关。本文就SIRT1在心血管疾病中的作用予以综述,为研究心血管疾病的病理机制以及制订临床防治方案提供新的思路。
SIRT1 is a nicotinamide adenine dinucleotide (NAD+)-dependent deacetylating enzyme,whose physiological effects are achieved by deacetylation of target proteins.In the cardiovascular system,SIRT1 is involved in the regulation of mitochondrial function,oxidative stress,inflammation,apoptosis and autophagy.The abnormality of SIRT1 activity is closely related to the occurrence and development of many cardiovascular diseases,such as heart failure,ischemia-reperfusion injury,myocardial hypertrophy,diabetic cardiomyopathy,atherosclerosis,and so on.This paper summarized the role of SIRT1 in cardiovascular diseases to provide new ideas for the pathological mechanism of cardiovascular diseases and clinical therapies.
