泽泻不同炮制品中7个化学成分的含量测定 被引量:5

摘要 目的:建立HPLC法测定不同方法炮制的泽泻中泽泻醇A、泽泻醇B、23-乙酰泽泻醇C、23-乙酰泽泻醇B、24-乙酰泽泻醇A、泽泻烯醇、环氧泽泻烯的含量,阐述不同炮制方法对泽泻中7个成分含量的影响。方法:采用HPLC法,色谱柱为Waters XTERRA C_(18)柱(250 mm×4.6 mm, 5μm),流动相为乙腈-0.01%磷酸水溶液,梯度洗脱,流速为1.0 ml·min^(-1),检测波长为208 nm、246 nm,柱温25℃。结果:泽泻醇A、泽泻醇B、23-乙酰泽泻醇C、23-乙酰泽泻醇B、24-乙酰泽泻醇A、泽泻烯醇、环氧泽泻烯线性范围分别为1.947~11.680μg·ml^(-1)(r=0.999 3)、1.756~10.540μg·ml^(-1)(r=0.999 2)、4.388~26.330μg·ml^(-1)(r=0.999 7)、8.420~50.520μg·ml^(-1)(r=0.999 8)、3.589~21.540μg·ml^(-1)(r=0.999 4)、1.476~8.854μg·ml^(-1)(r=0.999 5)、1.534~9.142μg·ml^(-1)(r=0.999 1);平均加样回收率分别为98.87%,98.72%,98.75%,98.85%,100.17%,99.20%,99.00%,RSD≤1.06%(n=6)。与生品相比,泽泻经炮制后23-乙酰泽泻醇C和23-乙酰泽泻醇B含量均降低,盐泽泻降幅最大;泽泻醇B、泽泻烯醇及环氧泽泻烯含量变化不明显;泽泻醇A、24-乙酰泽泻醇A的含量均增加。结论:该方法操作简单、结果准确、重复性好,可同时测定泽泻不同炮制品中7个化学成分的含量。 Objective: To establish an HPLC method for content determination of alisol A, alisol B, alisol C 23-acetate, alisol B 23-acetate, alisol A 24-acetate, alisonenol and alismoxide in different processed Alismatis Rhizoma, and to explain the effects of different processing methods on the seven components in Alismatis Rhizoma. Methods: The HPLC analysis was performed on a Waters XTERRA C_(18)column(250 mm×4.6 mm, 5 μm). The mobile phase was a mixture of acetonitrile-0.01% phosphoric acid with gradient elution. The flow rate was 1.0 ml·min;, the detection wavelengths were 208 and 246 nm, and the column temperature was set at 25 ℃. Results: The linear ranges of alisol A, alisol B, alisol C 23-acetate, alisol B 23-acetate, alisol A 24-acetate, lisonenol and alismoxide were 1.947-11.680 μg·ml^(-1)(r=0.999 3), 1.756-10.540 μg·ml^(-1)(r=0.999 2), 4.388-26.330 μg·ml^(-1)(r=0.999 7), 8.420-50.520 μg·ml^(-1)(r=0.999 8), 3.589-21.540 μg·ml^(-1)(r=0.999 4), 1.476-8.854 μg·ml^(-1)(r=0.999 5) and 1.534-9.142 μg·ml^(-1)(r=0.999 1), respectively. The average recoveries were 98.87%,98.72%,98.75%,98.85%,100.17%,99.20% and 99.00%,respectively. The RSDs were no more than 1.06%(n=6). Compared with the raw Alismatis Rhizoma, the contents of alisol C 23-acetate and alisol B 23-acetate were decreased after processing, and the decrease range of alisol salt was the widest;the contents of alisol B, alisonenol and alismoxide did not change significantly;the contents of alisol A and alisol A 24-acetate were increased. Conclusion: The method is simple, accurate and reproducible, which can be used for the simultaneous determination of the seven chemical components in different processed products of Alismatis Rhizoma.
