摘要
采用单因素及正交试验方差分析法优化长白山道地紫苏黄酮的提取条件,并初步研究其对小鼠的非特异性免疫功能的影响。结果表明:长白山紫苏黄酮的最佳提取条件为乙醇浓度70%、料液比1∶20(g/mL)、提取温度50℃、超声时间35 min,在此条件下长白山紫苏黄酮的得率为5.01%;通过小鼠碳粒廓清实验发现,长白山紫苏黄酮中剂量和高剂量组可显著、极显著提高小鼠碳粒廓清指数K和吞噬指数α,且高剂量组可促进小鼠胸腺及脾脏生长发育,使免疫器官指数升高,增强小鼠的非特异性免疫功能,表明长白山紫苏黄酮具有良好的免疫活性。
Conditions for extraction of flavonoids from the flavone content of Changbai Mountain Perilla were optimized by single factor and orthogonal test analyses of variance.Optimum extraction conditions were 70%ethanol,and a solid-liquid ratio of 1∶20(g/mL).Optimum extraction temperature was 50℃,and optimal ultrasonication time was 35 min.Under these conditions,the flavonoid yield from Changbai Mountain Perilla was 5.01%.A carbon particle clearance test in mice revealed that medium and high dose Changbai Mountain Perilla significantly increased the carbon clearance index K,and the phagocytic indexα.High dose Changbai Mountain Perilla extract could also promote growth and development of thymus and spleen,increase immune organ indices,and enhance non-specific immune function in mice,indicating that the flavonoids from Changbai Mountain Perilla stimulated good immune activity.
