摘要
建立了高效液相色谱--示差折光检测器测定鱼油甘油酯含量的方法。选用Agilent ZORBAX RX-SIL色谱柱(4.0mm×250mm,5μm),正己烷和异丙醇为流动相(15∶1,v∶v),流速0.8mL/min,柱温34oC,示差折光检测器(RID)检测。结果表明,油酸甘油三酯和油酸甘油一酯在5.0-25.0mg/mL范围内,方法的线性关系良好,相关系数分别为0.9999和0.9998;加标回收率分别为97.35%-100.47%、98.32%-99.65%,相对标准偏差(RSD)分别为1.59%、0.68%。该方法操作简便、精密度和方法重复性好,适用于鱼油甘油酯含量测定。
A method for determination of glycerol ester in triglycerol fish oil by high performance liquid chromatography(HPLC)with differential refractive index detector was established. The samples were analyzed by HPLC using Agilent ZORBAX RX-SIL column(4.0 mm×250 m,5μm)with(RID)detector. The mobile phase was a mixture of n-heptane and isopropanol(15∶1,v/v)at a flow rate of 0.8 mL/min. The results showed that a good linear relationship were obtained between the mass concentration of oleic acid triglyceride and oleic acid monoglyceride from 5.0 mg/mL to 15.0 mg/mL,and the correlation coefficients were 0.9999 and 0.9998,respectively;Average recovery rates of standard addition and relative standard deviations for Oleic acid triglyceride and Oleic acid monoglyceride were 97.35% - 100.04% and 98.32% - 99.65%,1.59% and 0.68%.The method is simple,accurate and reproducible,and has higher accuracy,suitable for the determination of glycerol ester in triglycerol fish oil.
