裸鼠脑胶质瘤原位移植模型的建立

摘要 目的建立脑胶质瘤原位移植模型。方法将荧光素酶标记的LN229细胞接种于裸鼠脑右侧尾状核区,术后对伤口进行消毒,保证裸鼠存活率。每周使用小动物活体成像仪监测肿瘤。实验结束,取出脑组织进行病理学检查。结果裸鼠脑内注射体外培养的LN229人脑胶质瘤细胞,注射后14 d,活体成像观察,每只裸鼠脑部均察见肿瘤生长,其荧光信号值均值达到8.93×10^(7);组织病理学检查符合人脑胶质瘤形态特征,未见脑外转移。结论本实验采用脑立体定位仪原位接种脑胶质瘤细胞可建立脑胶质瘤原位移植模型,此方法建立的模型均一性、成瘤率和存活率高,可作为评价药物抗脑胶质瘤效果的理想模型。 Objective To establish orthotopic transplantation model of glioma.Methods Luciferase labeled LN229 cells were inoculated into the right caudate nucleus of the brain of nude mice.After operation,the wound was disinfected to ensure the survival rate of nude mice.Small animal imagers were used to monitor tumors every week.At the end of the experiment,the brain tissue was taken out for pathological examination.Results LN229 human glioma cells cultured in vitro were injected into the brain of nude mice.14 days after injection,tumor growth was observed in the brain of each nude mouse,and the average value of fluorescence signal was 8.93×10^(7).Histopathological examination conformed to the morphological characteristics of human glioma and no extracerebral metastasis was found.Conclusion The model of orthotopic transplantation of glioma was established by orthotopic inoculation of glioma cells with brain stereotactic instrument in this study.The model established by this method has high homogeneity,high tumor formation rate and survival rate,and can be used as an ideal model to evaluate the effect of drugs against glioma.
