丹参灵芝三七茶辅助降血脂作用及安全性研究

Study on the blood lipid-lowering effect and safety of Danshen Ganoderma Notoginseng Tea

目的筛选丹参灵芝三七茶冲泡服用工艺参数,并考察其降血脂作用和安全性。方法以浸膏得率为指标,进行L_(9)(3^(4))正交试验设计,分析冲泡服用的最佳工艺参数。建立高血脂大鼠模型,以受试样品的中、高剂量灌胃42 d,同时设立煎煮液、空白和模型对照组,测定实验动物血脂水平。采用急性经口毒性试验、Ames试验、小鼠精子畸形试验、小鼠骨髓嗜多染红细胞微核试验、大鼠30 d喂养试验评价样品的食用安全性。结果冲泡服用最佳工艺条件为10倍加水量90℃、60 min提取2次;与模型对照组比较,样品高剂量组及煎煮液对照组大鼠血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)含量均显著下降(P<0.05),各剂量组血清高密度脂蛋白胆固醇(HDL-C)含量均无统计学意义(P>0.05);急性毒性试验结果显示样品属无毒级,未发现遗传毒性,大鼠30 d喂养试验未发现有意义的病理性损伤。结论丹参灵芝三七茶具有辅助降血脂作用,样品安全无毒,可长期服用。

Objective To screen the technological parameters of brewing and taking of Danshen Ganoderma Notoginseng Tea and its hypolipidemic effect and safety were investigated.Methods Taking the extract yield as the index,the L_(9)(3^(4))orthogonal experiment design was carried out to investigate the best technological parameters of brewing and taking of Danshen Ganoderma Notoginseng Tea.A hyperlipidemic rat model was established,and the test group animals were administered intragastrically to the test samples at medium dose(10 times)and high(30 times)doses for 42 days.At the same time,a decoction control group,a blank control group and a model control group were set up.The blood lipid levels of the animals in each group were determined.The acute oral toxicity test,Ames test,mouse sperm deformity test,mouse bone marrow polychromatic erythrocyte micronucleus test and rat 30-day feeding test were used to evaluate the safety of Danshen Ganoderma Notogingseng Tea.Results The optimal process conditions:the number of extractions was 2 times,the extraction time was 60 minutes,the amount of water added was 10 times,and the extraction temperature was 90℃.The results showed that compared with the model control group,the serum TC,LDL-C and TG contents of the high dose group and the decoction control group decreased significantly(P<0.05),but the HDL-C contents of each dose group had no statistical significance(P>0.05).The results of the acute oral toxicity test showed that the sample was non-toxic.No significant pathological damage was found in the 30-day feeding test in rats.Conclusion Danshen Ganoderma Notoginseng Tea has the effect of assisting blood lipid lowering.The sample is safe and non-toxic and can be taken for a long time.
