白介素10基因rs1800896、rs1800871单核苷酸多态性与口腔鳞状细胞癌相关性研究

The Correlation between Single Nucleotide Polymorphisms of Interleukin 10 Gene rs1800896,rs1800871 and Oral Squamous Cell Carcinoma

目的:分析白介素10(interleukin-10,IL-10)基因rs1800896(-1082 A/G)、rs1800871(-819 T/C)位点与口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)疾病相关性。方法:选取125例OSCC患者(病例组)及110例性别、年龄均匹配的外伤患者(对照组),提取研究对象基因组DNA进行PCR反应后将产物直接测序,比较并分析-1082 A/G和-819 T/C各基因型在两组间的分布频率。结果:关联研究结果显示-819 T/C位点TC基因型人群较TT基因型人群、CC基因型人群较TT+TC基因型人群,以及C等位基因型人群OSCC发病风险显著增加;Logistic分层分析显示在男性、年龄≥60岁、饮酒及不吸烟人群中OSCC发病风险也有所增加。结论:IL-10基因-819 T/C位点变异与中国汉族人群OSCC疾病发生存在相关性,同时长期吸烟与饮酒可能增加OSCC发病风险。

Objective To analyze the relationship between rs1800896(-1082 A/G)and rs1800871(-819 T/C)of inter⁃leukin-10(IL-10)gene and oral squamous cell carcinoma(OSCC).Methods A total of 125 patients with OSCC tumors were selected as the case group,and 110 patients with gender and age matching trauma were selected as the control group.Genomic DNA of the subjects was extracted for PCR reaction,and the products were directly sequenced.The distribution frequencies of-1082 A/G and-819 T/C genotypes between the two groups were compared and analyzed.Results The re⁃sults of association study showed that the risk of OSCC was significantly increased in-819 T/C TC genotype carriers com⁃pared with TT genotype and in CC genotype carriers compared with TT+TC genotype and C allele carriers.Logistic stratified analysis showed that the risk of OSCC was also increased in men,age≥60,drinking and non-smoking.Conclusion There is a correlation between the mutation of IL-10 gene-819 T/C locus and the occurrence of OSCC in Han Chinese population.Meanwhile,long-term smoking and drinking may increase the risk of OSCC.
