miRNA-720、miR-17-3p及miR-215-5p在多发性骨髓瘤中的表达及临床意义

Expression and clinical significance of miRNA-720,miR-17-3p and miR-215-5p in multiple myeloma

目的:探讨miRNA-720、miR-17-3p及miR-215-5p在多发性骨髓瘤(MM)中的表达及临床意义。方法:提取100例MM患者(病例组)和100名健康人群(对照组)血清RNA。实时荧光定量PCR测定血清miRNA-720、miR-17-3p及miR-215-5p表达水平,分析其表达量与临床病理特征的关系,Kaplan-Meier生存分析法分析三种不同基因相对表达量与患者预后的关系。结果:病例组血清miRNA-720、miR-17-3p相对表达量高于对照组(P<0.05),miR-215-5p相对表达量低于对照组(P<0.05);血清miR-17-3p相对表达量与ISS分期有统计学差异(P<0.05),血清miR-215-5p相对表达量与血红蛋白水平有统计学差异(P<0.05);血清miRNA-720高表达患者初次化疗有效率低于低表达患者(P<0.05),血清miR-215-5p低表达患者初次化疗有效率低于高表达患者(P<0.05);miRNA-720及miR-17-3p低表达和miR-215-5p高表达患者生存情况较好(P<0.05)。结论:miRNA-720、miR-17-3p及miR-215-5p可能与MM密切相关,可作为MM诊断、治疗及预后的评估指标。

Objective:To analyze the expression and clinical significance of miRNA-720,miR-17-3p and miR-215-5p in multiple myeloma(MM).Methods:Serum RNA was extracted from 100 mm patients(case group)and 100 healthy people(control group).The expression levels of miRNA-720,miR-17-3p and miR-215-5p in serum were determined by real-time quantitative PCR.The relationship between the expression levels and clinicopathological features was analyzed.The relationship between the relative expression levels of three different genes and the prognosis of patients was analyzed by Kaplan Meier survival analysis.Results:The relative expression levels of serum miRNA-720 and miR-17-3p in the case group were significantly higher than those in the control group(P<0.05),and the relative expression levels of miR-215-5p in the case group were significantly lower than those in the control group(P<0.05).There was significant difference between the relative expression of serum miR-17-3p and ISS stage(P<0.05),and the relative expression of serum miR-215-5p was significant different from hemoglobin level(P<0.05).The response rate of primary chemotherapy in patients with high expression of serum miR-720 was lower than that in patients with low expression(P<0.05),and the response rate of primary chemotherapy in patients with low expression of serum mR-215-5p was lower than that in patients with high expression(P<0.05).The patients with low expression of miRNA-720,low expression of miR-17-3p and high expression of miR-215-5p had better survival(P<0.05).Conclusion:MiRNA-720,miR-17-3p and miR-215-5p may be closely related to MM and can be used as an indicator for the diagnosis,treatment and prognosis of MM.
