酸碱指示剂在工业微生物高通量筛选中的应用进展 被引量:1

摘要 随着工业微生物筛选通量的逐级提高,高通量筛选技术作为新型、自动化、高灵敏度和大规模集群式筛选体系被广泛应用。目前,作为高通量筛选体系关键步骤的高通量检测方法主要有质谱法、酸碱指示剂法、特殊光谱法、免疫法法、紫外光/可见广谱法和荧光信号法等,其中酸碱指示剂法因具有成本低、筛选体系小、范围广、速度快和原理简单的优势成为研究热点。酸碱指示剂通过反应体系中质子的变化显色,显色吸光度与pH具有良好的线性相关性,广泛应用于功能微生物的初筛、酶活检测和代谢产物检测等领域。主要对酸碱指示剂的应用、选择原则和指示剂种类的研究现状进行了概述,探讨了酸碱指示剂筛选方法在工业微生物高通量筛选中的重要性和未来的研究趋势。 With the gradual increase in the screening throughput of industrial microorganisms,high throughput screening has widely used as a new,automated,highly sensitive and large-scale cluster screening system.At present,high throughput analytical technologies,such as mass spectrometry,acid-base indicator method,spectroscopy,immunization,ultraviolet/visible broad-spectrum method and fluorescence,are the key steps of screening system.Among them,acid-base indicator method has become a research hotspot due to its advantages of low cost,small screening system,wide range,simple principle,and efficiency.The acid-base indicator develops color through the change of protons in the reaction system,and the color absorbance has a good linear correlation with pH.Then acid-base indicator is widely used in primary screening of industrial microorganism,enzyme activity assay,metabolites detection and other fields.This paper mainly summarized the research status of acid-base indicator application,choice principle and indicator types,and discussed the importance of acid-base indicator in high throughput screening of industrial microorganisms,as well as the future research trends.
