摘要
为了实现多杀菌素工业化生产,本次试验进行了多杀菌素发酵从摇瓶到50L发酵罐放大的研究,试验从摇瓶种子液培养放大到20L种子罐种子液培养,找出了最佳适合孢子发芽菌丝繁殖的10号培养基,淀粉15g/L、酵母膏3g/L、酪蛋白30g/L、KH_(2)PO_(4)0.5g/L、MgSO_(4)2g/L,并且经过优化筛选出更经济实用国产原材料胰蛋白替代价格较高的酪蛋白;从摇瓶发酵液培养放大到50L发酵罐培养,通过控制菌丝体生长对数期的搅拌转速,由迟滞期200r/min调整到500r/min,加速菌体从初级代谢快速进入次级代谢,保证发酵液的pH由最高点7.4以上,快速降到7.0以下,菌丝体进入到生产期;优化改进50L罐发酵使用的碳源,筛选出最适合使用发酵单一碳源葡萄糖,避免引起葡萄糖效应,确定了基础培养基最佳葡萄糖含量8%以下;确定不论是何种消泡剂发酵液中含量0.1%(含0.1%)以上都对多杀菌素发酵产物合成不同程度的影响,基础培养基的添加料应控制在0.015%-0.02%,发酵过程采用一次滴加的方法加入,整个发酵过程减少消泡剂的加入量。
In order to realize the industrial production of polyfungicide,This test conducted polybactericidal fermentation from rocker to 50L fermenter amplification.The experiment was amplified from shaking bottle seed fluid culture to 20L seed tank seed culture,found the#10 medium best suitable for spore germination mycelium reproduction,starch 15g/L,yeast cream 3g/L,casein 30g/L,KH_(2)PO_(4)0.5g/L,MgSO_(4)2g/L,And after optimization of more economical and practical domestic raw materials trypsin to replace the higher price of casein;Expanding from bottling fermentation fluid culture to 50L fermenter culture,by controlling the mixing speed of mycelium growth in logarithmic periods,from hysteresis 200r/min to 500r/min,accelerated bacterial body with rapid entry into secondary metabolism from primary metabolism,ensure that the pH of the fermentation fluid is above 7.4,fast descent to below 7.0,mycelium enters the production period;Optimize improved carbon sources for 50L tank fermentation,screening out the only carbon source glucose most suitable for fermentation,avoid causing the glucose effect,optimum base medium glucose content was determined below 8%;Determine the influence of the fermentation liquid content above 0.1%(including 0.1%)on the synthesis of polyfungicide fermentation products,adaddition of base medium shall be controlled at 0.015%-0.02%The fermentation process is added by the primary drip method,the entire fermentation process reduces the amount of antifoaming agent.
