基于网络药理学探讨玉屏风散治疗系统性红斑狼疮的作用机制 被引量:2

摘要 目的采用网络药理学的方法研究方剂玉屏风散的主要活性成分、靶点,初步探讨其治疗系统性红斑狼疮的作用机制。方法通过TCMSP、Uniprot数据库筛选出玉屏风散的药物活性靶点,通过OMIM、GeneCards、TTD数据库筛选出系统性红斑狼疮疾病相关靶点信息。采用RStudio软件将玉屏风散的药物活性靶点与系统性红斑狼疮疾病靶点信息取交集,将交集靶点数据依靠String在线数据库平台构建蛋白互作PPI网络模型,利用Cytoscape 3.7.2软件,构建可视化的“中药-成分-靶点-疾病”网络图并进行分析,找寻玉屏风散治疗系统性红斑狼疮的关键靶点并通过RStudio绘制柱状图。使用RStudio对关键靶点进行GO功能分析和KEGG通路分析。结果玉屏风散治疗系统性红斑狼疮的潜在靶点有124个,蛋白互作PPI网络模型提示AKT1、VEGFA、IL6、TNF、TP53等可能是玉屏风散治疗系统性红斑狼疮的核心靶点。GO功能分析结果显示,玉屏风散治疗系统性红斑狼疮潜在基因的生物功能主要涉及细胞因子受体结合、细胞因子活性、受体配体活性等;KEGG通路分析结果显示,玉屏风散治疗SLE潜在基因的通路主要涉及糖尿病并发症中的AGE-RAGE信号通路、IL-17信号通路、肿瘤坏死因子信号通路等信号通路。结论通过网络药理学分析表明方剂玉屏风散通过多个靶点调控多条信号转导通路发挥对系统性红斑狼疮的治疗作用,为玉屏风散的进一步研究和应用奠定了基础。 Objective To study the main active ingredients and targets of the prescription YuPingFengSan by network pharmacology,and to preliminarily explore its mechanism of action in the treatment of systemic lupus erythematosus.Methods The prescription drug active targets of YuPingFengSan were screened through TCMSP and Uniprot databases,and the target information related to systemic lupus erythematosus disease was screened through OMIM,GeneCards,and TTD databases.RStudio software was used to collect the information of drug active targets of YuPingFengSan and SLE disease target.String online database platform constructed the PPI network model of protein interaction relationship.Using the software of Cytoscape3.7.2,the visualized network diagram of"Chinese Medicine-components-targets-diseases"was constructed and analyzed to find the key targets of YuPingFengSan in the treatment of SLE and draw a histogram through RStudio.Rstudio software was used to analyze the GO function and KEGG pathway of key targets.Results There were 124 potential targets of YuPingFengSan in the treatment of SLE.The PPI network model suggests that AKT1,VEGFA,IL6,TNF,TP53,etc.may be the core targets of YuPingFengSan in the treatment of SLE.The results of GO function analysis showed that the biological functions of YuPingFengSan in the treatment of SLE potential genes mainly involve cytokine receptor binding,cytokine activity,receptor ligand activity,etc.;KEGG pathway analysis results show that YuPingFengSan’s pathway for treating SLE potential genes mainly involves in the AGE-RAGE signaling pathway in diabetic complications,IL-17 signaling pathway,tumor necrosis factor signaling pathway,TNF signaling pathway and other signaling pathways.Conclusion In this study,we found that,by the network pharmacology analysis,the prescription YuPingFengSan regulates multiple signal transduction pathways through multiple targets to exert its therapeutic effect on SLE,which laid the foundation for the further research and application of YuPingFengSan.
