不同酶对巫山淫羊藿中淫羊藿苷和朝藿定C含量的影响及抑菌活性研究

Effects of Different Enzymes on Contents of Icariin and Epimedin C from Epimedium wushanense Ying and Bacteriostatic Activity

摘要 在巫山淫羊藿提取物中分别加入α-淀粉酶、果胶酶、纤维素酶进行酶解,经大孔树脂纯化后采用HPLC法测定巫山淫羊藿中淫羊藿苷和朝藿定C含量;并初步研究了巫山淫羊藿提取物对金黄色葡萄球菌和大肠杆菌的抑菌活性。结果表明,巫山淫羊藿提取物在经过酶解和纯化后,淫羊藿苷和朝藿定C的含量均明显增加,其中果胶酶对两者含量的影响最大,巫山淫羊藿提取物在经过果胶酶酶解和大孔树脂纯化后,淫羊藿苷和朝藿定C的含量分别增至11.65%、31.79%;巫山淫羊藿提取物及果胶酶酶解物对金黄色葡萄球菌和大肠杆菌均有较好的抑菌效果,但未酶解的巫山淫羊藿提取物的抑菌活性优于酶解物和纯化物。 We determined the contents of icariin and epimedin C in Epimedium wushanense Ying through content determination by HPLC method after enzymolysis withα-amylase,pectinase,and cellulase and purification by macroporous resin.Moreover,we preliminarily studied the bacteriostatic activity of Epimedium wushanense Ying extract against Staphylococcus aureus and Escherichia coli.The results show that after enzymolysis and purification of Epimedium wushanense Ying extract,the contents of icariin and epimedin C significantly increase to 11.65%and 31.79%,respectively,and pectinase has the greatest effect on their contents.The Epimedium wushanense Ying extract and pectinase hydrolysate have good bacteriostatic effects on Staphylococcus aureus and Escherichia coli,but the bacteriostatic activity of Epimedium wushanense Ying extract without enzymolysis is superior to that of the hydrolysate and the purified product.
