谷氨酸、γ-氨基丁酸在偏头痛模型PAG和TNC中的变化

摘要 目的探讨偏头痛模型中脑导水管周围灰质(PAG)和三叉神经脊束核尾侧亚核(TNC)中的Glu、GABA含量变化。方法30只SD雄性大鼠随机分为对照组、模型组和西药组,每组各10只。对照组、模型组灌胃超纯水,西药组灌胃利扎曲普坦,连续灌胃7天后,对照组腹腔注射生理盐水,模型组、西药组腹腔注射硝酸甘油。然后,进行福尔马林试验。最后,从脑组织中分离出PAG与TNC,进行离体磁共振波谱扫描c结果PAG内的cGABA(Coefficient=0.261)、cGlu(Coefficient=0.146)、rGlu(Coefficient=0.040)均高于RTNC(Coefficient=0),差异有统计学意义(P<0.01)。模型组PAG内的rGlu(0.57±0.05)较对照组(0.65±0.07)减少(P=0.031)。西药组PAG内的rGlu[0.62(0.57,0.66)]与模型组(0.57±0.05)比较有增多趋势(P=0.288)。结论PAG和TNC内Glu含量不同。大鼠在注射硝酸甘油后,福尔马林诱导的PAG中Glu释放减少。 Objective To investigate the changes of Glu and GABA content in periaqueductal gray(PAG)and caudal spinal trigeminal nucleus(TNC)in migraine model.Methods Thirty male SD rats were randomly divided into control group,model group and western medicine group,with 10 rats in each group.The control group and model group were intragastrically given ultra-pure water,and the Western medicine group was intragastrically given rizatriptan.After continuous intragastrical administration for 7 days,the control group was intraperitoneally injected with normal saline,and the model group and western medicine group were intraperitoneally injected with nitroglycerin.Then,formalin test was performed.Finally,PAG and TNC were separated from brain tissue for in vitro magnetic resonance spectrum scanning.Results CGABA(Coefficient=0.261),cGlu(Coefficient=0.146)and rGlu(Coefficient=0.040)in PAG were higher than those in RTNC(Coefficient=0),and the differences were statistically significant(P<0.01).The rGlu in model group(0.57±0.05)was lower than that in the control group(0.65±0.07)(P=0.031).The rGlu in PAG of western medicine group[0.62(0.57,0.66)]was higher than that of model group(0.57±0.05)(P=0.288).Conclusion The content of Glu is different in PAG and TNC.Formalin-induced PAG Glu release was reduced in rats after nitroglycerin injection.
