摘要
目的观察骨骼肌缺血再灌注后大鼠血清炎性指标改变及磷脂酶Cδ3(PLCδ3)表达水平变化,探究磷脂酶Cδ3在骨骼肌缺血再灌注中的作用。方法以120只大鼠为研究对象,随机分为4组建立骨骼肌缺血再灌注模型(对照2小时组,对照4小时组,缺血2小时组,缺血4小时组),每组各30只大鼠,在大鼠缺血再灌注1天、3天和7天后分别检测各组大鼠血清IL-6、IL-1β含量水平及骨骼肌中PLCδ3表达水平。结果缺血再灌注大鼠血清中的炎性指标显著增加,同时缺血4小时组大鼠再灌注1天、3天和7天后血清中IL-6和IL-1β含量均显著高于缺血2小时组水平(P<0.05),而PLCδ3表达水平低于缺血再灌注2小时组。结论PLCδ3表达水平在一定范围内与骨骼肌损伤水平趋势相同,有望成为新的缺血再灌注损伤治疗靶点。
Objective To observe the changes of serum inflammatory indexes and phospholipase Cδ3(PLCδ3)expression in rats after skeletal muscle ischemia-reperfusion,and to investigate the role of phospholipase C in skeletal muscle ischemia reperfusion.Methods 120 rats were randomly divided into 4 groups(control group 2h,control group 4h,ischemia group 2h,ischemia group 4h)to establish skeletal muscle ischemia-reperfusion model,with 30 rats in each group.The levels of IL-1β,IL-6 in serum and PLCδ3 in skeletal muscle of rats in each group were detected 1d,3d and 7d after ischemia-reperfusion.Results The inflammatory indexes in serum of ischemia-reperfusion rats were significantly increased.Meanwhile,the serum levels of IL-6 and IL-1βin 4h group were significantly higher than those in 2h group after reperfusion at 1d,3d and 7d(P<0.05),while the expression level of PLCδ3 was lower than that in 2h group.Conclusion The level of PLCδ3 is similar to the level of skeletal muscle injury in a certain range,which is expected to be a new therapeutic target for ischemia-reperfusion injury.
