狭山野豌豆总黄酮富集工艺研究

摘要 目的:对狭山野豌豆总黄酮的富集工艺进行研究.方法:实验以静态吸附和静态解吸附为考察指标,对10种不同型号的大孔树脂进行筛选,优选出对狭山野豌豆总黄酮具有较好吸附和分离性能的树脂,并进一步对该树脂进行纯化工艺考察.结果:优选出的大孔树脂型号是HPD-826,上样质量浓度为0.1 g/mL,用1 mol/mL HCl将样品水溶液的pH调节为5~6,上样体积为5倍树脂柱床体积,当调节流速为2 mL/min时进行动态吸附,洗脱剂为70%乙醇水溶液,以3 mL/min流速进行洗脱,洗脱体积为10倍树脂床体积数.通过HPD-826型大孔树脂对狭山野豌豆总黄酮富集,结果为3.28960 mg/g.结论:HPD-826型大孔树脂适于对狭山野豌豆总黄酮的富集,富集量好于乙酸乙酯萃取的方法. Objective:To study the extraction and purification process of total flavonoids in Vicia amoena Fisch.Var.angusta Freyn.Method:In this experiment,static adsorption and static desorption were used as the investigation indexes to screen the 10 types of macroporous resins,and the resin with better adsorption and separation performance for total flavonoids was optimized,and the purification process of the resin was further investigated.Results:The preferred macroporous resin model is HPD-826.The sample concentration was 0.1 g/mL,adjust the pH of the sample aqueous solution to 5~6 with 1 mol/mL HCl,the sample volume was 5 times the volume of the resin column bed,the flow rate was adjusted to 2 mL/min for dynamic adsorption,the eluent was 70%ethanol aqueous solution,and the elution volume was 3 mL/min,and the elution volume was 10 times the volume of the resin bed.The total flavonoids were enriched by HPD-826 macroporous resin,and the result was 3.28960 mg/g.Conclusion:HPD-826 macroporous resin was suitable for the enrichment of total flavonoids in Vicia amoena Fisch.Var.angusta Freyn,and the concentration is better than that of ethyl acetate extraction.
