基于网络药理学探讨复方苦参汤治疗溃疡性结肠炎的作用机制 被引量:2

The mechanism of action of Compound Sophora decoction in the treatment of ulcerative colitis: analysis based on the network pharmacology of traditional Chinese medicine

摘要 目的应用网络药理学研究复方苦参汤治疗溃疡性结肠炎的物质基础及其可能的作用机制。方法应用TCMSP数据库挖掘复方苦参汤潜在活性成分及其药物靶点,通过GeneCards、DrugBank、DisGeNet、TherapeuticTargetDatabase数据库筛选疾病靶点。将复方苦参汤活性成分的药物靶点与疾病靶点匹配,所得交集靶点导入String构建PPI网络,借助Cyto NCA插件计算节点信息,经拓扑分析,筛选关键靶点。使用Cytoscape构建可视化"药物-靶点-疾病"网络并进行分析。使用DAVID数据库对关键靶点进行GO和KEGG富集分析。结果复方苦参汤的潜在活性药物成分有233个,包括槲皮素、木樨草素、槐异二氢黄酮A等。药物与疾病的交集靶点有180个,治疗溃疡性结肠炎的关键靶点为AKT1、MAPK1、MAPK3、STAT3、JUN、HSP90AA1等。主要发挥作用的信号通路为:癌症信号通路、HIF-1信号通路、TNF信号通路、PI3K-AKT信号通路等。结论复方苦参汤可通过多靶点、多通路、多途径治疗溃疡性结肠炎。 Objective To study the material basis and possible mechanism of Compound Sophorae Decoction in the treatment of Ulcerative colitis(UC) by using network pharmacology.Methods The active components of Compound Sophorae Decoction were collected by TCMSP database to collect the potential targets of active components, the disease targets related to ulcerative colitis were collected by GeneCards database, DrugBank, DisGeNET database and TherapeuticTargetDatabase.The drug targets of the active ingredients of Compound Sopranthe decoction were matched with the disease targets, and the obtained intersection targets were imported into String to construct PPI network.Visual drug-target-disease networks were constructed and analyzed using Cytoscape.GO and KEGG enrichment analysis of key targets was performed using David database.Results There were 233 potential active drug components in Compound Sophora decoction, including quercetin, luteolin, isohydroflavone A, etc.There are 180 intersection targets between drugs and diseases, and the key targets for the treatment of UC are Akt1, MAPK1, MAPK3, Stat3, Jun, Hsp90 AA1, etc.The main signaling pathways are pathway in cancer, HIF-1 signaling pathway, TNF signaling pathway, PI3 K-AKT signaling pathway and so on.Conclusion Compound Sophora decoction can treat ulcerative colitis through multi-target, multi-channel and multi-pathway.
