PyHY5基因遗传转化矮牵牛的条件筛选与优化

摘要 本研究以矮牵牛‘轻浪’(Easy wave)和‘超级小瀑布’(Supercascade)两个不同品种的无菌苗叶片作为试验材料,采用叶盘转化法进行试验。设置不同的侵染时间(5,10,15,20 min)和潮霉素浓度(5,10,15 mg/L),培养40 d,统计矮牵牛叶片的转化率,筛选出PyHY5基因遗传转化矮牵牛的适宜条件。结果表明,‘轻浪’的转化率较高,侵染时间为10 min、潮霉素的浓度为10 mg/L时,最终的遗传转化效率为5%,适合作为矮牵牛的遗传转化条件,这为Py HY5基因的进一步研究提供参考。 Two different varieties of sterile seedling leaves of’Easy wave’and’Supercascade’of Petunia hybrida Vilm were used as the test materials,and the leaf disk transformation method was used.Setting different infection time(5,10,15,20 min),hygromycin concentration(5,10,15 mg/L)and culture for 40 days,the conversion rate of Petunia hybrida leaves was counted,and the suitable conditions for PyHY5 gene genetic transformation were screened out.The results showed that the concentration rate of’easy wave’was relatively high.When the infection time was 10 min and the concentration of hygromycin was 10 mg/L,the final genetic conversion efficiency was 5%,which was suitable as a genetic transformation condition for Petunia hybrida leaves.This result can provides reference for further research on Py HY5 gene.
