近红外二区荧光探针的设计及应用研究进展

摘要 高时空分辨率和高灵敏度的荧光成像技术是一种新兴的活体可视化检测工具.与近红外一区(NIR-Ⅰ,700~900 nm)相比,近红外二区(NIR-Ⅱ,1000~1700 nm)成像具有更低的自发背景荧光、更深的组织穿透性和更高的信背比,因此NIR-Ⅱ荧光成像能促进深部疾病的精确诊断.NIR-Ⅱ荧光探针是NIR-Ⅱ荧光成像的基础.目前已开发一系列基于有机和无机材料的NIR-Ⅱ荧光探针,包括有机小分子染料、基于小分子染料的有机纳米粒子、共轭聚合物、量子点、稀土掺杂纳米粒子和单壁碳纳米管等.本文综述近期新型NIR-Ⅱ荧光探针的研究进展及其在生物医学领域的应用. Fluorescence imaging with a high spatial-temporal resolution and high sensitivity provides a powerful tool for visualizing dynamic biological processes.Compared with the first near-infrared window(NIR-Ⅰ,700-900 nm),the second near-infrared window(NIR-Ⅱ,1000-1700 nm)imaging shows lower spontaneous background fluorescence,deeper tissue penetrability and a higher signal-to-background ratio.Therefore,NIR-Ⅱfluorescence imaging could promote accurate diagnosis of deep diseases.The NIR-Ⅱfluorescent probe is the basis of NIR-Ⅱfluorescence imaging.At present,a series of NIR-Ⅱfluorescent probes based on organic and inorganic materials have been developed,including organic small molecule dyes,organic nanoparticles based on small molecule dyes,conjugated polymers,quantum dots,rare earth doped nanoparticles and single-walled carbon nanotubes.This paper reviews recent research progress in NIR-Ⅱfluorescent probe and its application in the biomedical field.
