秀珍菇多酚提取工艺优化及抗氧化活性探究 被引量:5

Optimization on Extraction Technology of Polyphenols from Pleurotus geesteranus and Study on the Antioxidant Activity

摘要 目的:应用响应面设计优化超声辅助水提秀珍菇多酚工艺,分析多酚的抗氧化能力。方法:应用单因素与Box-Behnken设计,优化秀珍菇多酚提取工艺,并建立多酚体外总抗氧化分析与DPPH、ABTS自由基清除分析。结果:优化设计实验结果表明秀珍菇多酚最佳提取工艺为:液料比68∶1(mL/g),超声功率300 W,提取时间87 min,提取温度45℃,多酚得率为(17.428±0.121)mg/g。体外抗氧化分析结果表明,秀珍菇多酚总抗氧化能力EC_(50)=0.177 mg/mL,DPPH自由基清除率与ABTS自由基清除率IC_(50)分别为0.363 mg/mL与0.008 mg/mL。结论:秀珍菇多酚的提取工艺可行,秀珍菇多酚具有较强的抗氧化能力。 Objective:Response surface design(RSM)is used to optimize the ultrasound-assisted water extraction process of polyphenols from Pleurotus geesteranus and to analyze the antioxidant capacity of polyphenols.Methods:Single factor and Box-Behnken design are used to optimize the extraction process of polyphenols from Pleurotus geesteranus,and total antioxidant analysis and DPPH and ABTS radicals scavenging analysis of polyphenols in vitro are established.Results:The optimal extraction conditions are as follows:liquid-solid ratio is 68∶1(mL/g),ultrasonic power is 300 W,extraction time is 87 min,extraction temperature is 45℃,and the yield of polyphenols is(17.428±0.121)mg/g.The results of antioxidant analysis in vitro show that the total antioxidant capacity EC_(50) is 0.177 mg/mL,the IC_(50) of DPPH radical scavenging rate and ABTS radical scavenging rate is 0.363 mg/mL and 0.008 mg/mL respectively.Conclusion:The extraction technology of polyphenols from Pleurotus geesteranus is feasible,and polyphenols from Pleurotus geesteranus have strong antioxidant ability.
