摘要
目的探讨hsa-miR-301a-3p对缺氧/复氧(hypoxia/reoxygenation, H/R)条件下重组组织型纤溶酶原激活剂(Recombinant tissue plasminogen activator, rtPA)相关的脑微血管内皮细胞死亡的作用。方法建立人体外脑微血管内皮细胞(Human brain microvascular endothelial cells, hBMECs)缺氧复氧模型,应用rtPA干预后检测细胞死亡数量及hsa-miR-301a-3p的改变,并进一步干预hsa-miR-301a-3p后再检测细胞死亡数量,随后在生信学分析的基础上进一步检测靶基因的变化。结果 rtPA干预可引起内皮细胞死亡数量增加及hsa-miR-301a-3p表达水平下降,hsa-miR-301a-3p模拟物(Mimic)可抑制rtPA相关的内皮细胞死亡,并引起下游靶基因缝隙连接蛋白43(Connexin43,Cx43)表达减少。结论 hsa-miR-301a-3p有助于减少缺氧复氧条件下rtPA相关的脑微血管内皮细胞死亡。
Objective To investigate the effects of hsa-miR-301 A-3 p on cell death after recombinant tissue plasminogen activator(rtPA) treatment in hypoxia/reoxygenation(H/R)conditions. Methods A hypoxic and reoxygenated model of human brain microvascular endothelial cells(HBMECs) was established. The number of cell death and hsa-miR-301 A-3 p expression were detected after rtPA intervention with or without has-mir-301 A-3 p interference, followed by further investigation of target genes on the basis of bioinformation analysis. Results The number of endothelial cell death and the expression of hsa-miR-301 A-3 p were increased after the intervention of rtPA. Hsa-miR-301 A-3 p mimic could inhibit the rtPA-related endothelial cell death and decrease the expression of downstream target gene connexin43(CX43) to decrease. Conclusion Hsa-miR-301 a-3 p can reduce rtPA-related brain microvascular endothelial cell death under hypoxia/reoxygenationconditions.
