摘要
目的研究促红细胞生成素(EPO)对大鼠剖宫产后创面愈合的促进作用及具体机制。方法将14 d成年雌性Spragve-Dawley孕鼠分为对照组(9只),EPO 10 U/kg组(9只),EPO 100 U/kg组(9只),EPO 1000 U/kg组(9只)。苏木素-伊红(HE)和PAS染色研究大鼠剖宫产后伤口愈合及组织形态变化;压力传感器和放大记录器检测EPO处理后伤口拉伸强度和子宫破裂压的变化;RT-qPCR检测伤口组织中转化生长因子-β1(TGF-β1)、成纤维生长因子(bFGF)、血小板-内皮细胞黏附分子(CD31)mRNA相对表达水平。结果与对照组比较,EPO 100 U/kg组和EPO 1000 U/kg组大鼠剖宫产后皮肤伤口拉伸强度和子宫破裂压显著增加(P<0.01)。HE和PAS染色结果显示,与对照组比较,EPO 100 U/kg组和EPO 1000 U/kg组子宫内膜腺体数量和子宫血管密度显著增加(P<0.05),伤口组织中羟脯氨酸的水平显著升高(P<0.05)。与对照组比较,EPO 100 U/kg组和EPO 1000 U/kg组伤口组织中TGF-β1、bFGF和CD31 mRNA表达明显升高(P<0.01)。结论EPO可通过增加TGF-β1和bFGF表达,减轻炎性反应并促进剖宫产后伤口愈合。
Objective To investigate the effect of erythropoietin(EPO)on wound healing after cesarean section in rats and its specific mechanism.Methods The adult female Sprague-Dawley pregnant rats at 14 days were randomly divided into the control group(Vehicle,9 rats),the EPO 10 U/kg group(9 rats),the EPO 100 U/kg group(9 rats),and the EPO 1000 U/kg group(9 rats).The HE and PAS staining were used to observe the wound healing and tissue morphology changes after cesarean section in rats;the pressure sensor and magnifying recorder were used to detect the changes in wound tensile strength and uterine rupture pressure after EPO treatment;the RT-qPCR was used to detect the hydroxyproline in the wound tissue and the mRNA expression levels of transforming growth factor-β1(TGF-β1),fibroblast growth factor(bFGF)and platelet-endothelial cell adhesion molecule(CD31).Results Compared with the control group,the skin wound tensile strength and uterine rupture pressure in the EPO 100 U/kg group and the EPO 1000 U/kg group increased significantly after the cesarean section in a dose-dependent manner(P<0.01).The HE and PAS staining results showed that compared with the control group,the number of endometrial glands and uterine vessel density in the EPO 100 U/kg group and the EPO 1000 U/kg group increased significantly.Compared with the control group,the TGF-β1,bFGF and CD 31 mRNA expression in the wound tissue of the EPO 100 U/kg group(9 rats)and the EPO 1000 U/kg group significantly increased(P<0.01).Conclusion EPO can promote wound healing after cesarean section by increasing the expression of the TGF-β1 and bFGF,and reducing the inflammatory reaction.
