摘要
目的:考察减压与常压提取对黄精糕降血糖活性成分的影响,从而优选一种科学、合理、稳定、可行的提取方法。方法:以黄精糕中降血糖活性成分和指标成分总黄酮、总多糖、葡萄糖、鞣花酸、山柰酚-3-O-芸香糖苷与干浸膏得率为评价指标,先用常压回流提取L_(9)(3^(4))正交设计试验法优化醇浓度、加溶剂量、提取时间、提取温度等常压与减压共有的提取工艺参数,再采用最佳工艺参数进行减压提取,95%可信区间重叠法比较减压与常压提取各成分的差异。结果:常压提取最佳工艺参数为:先用醇常压提取1次,加12倍量70%的乙醇,60℃提取1 h,再用水提2次,加水量分别为药材量的12、10倍,水提温度90℃,时间分别为2、1.5 h,减压提取醇浓度、加溶剂量、提取温度、提取时间皆同常压提取,仅提取压力为70%醇提150 hpa,水提70 hpa,减压提取醇提液中总黄酮、鞣花酸含量高于常压提取(P<0.05),减压提取水提液中葡萄糖含量高于常压提取(P<0.05)。结论:减压提取优于常压提取,减压提取最优工艺参数为70%醇提70 hpa,水提150 hpa,先用70%乙醇减压提取1次,加入12倍量70%的乙醇,60℃减压提取1 h,再用水减压提取2次,加水量分别为药材量的12、10倍,水提取温度90℃,时间分别为2、1.5 h。减压提取工艺科学、合理、稳定、可行,可为工业生产提供科学依据。
【Objective】To investigate the effect of decompression and atmospheric pressure extraction on the hypoglycemic active components of Huangjing cake,and to select a scientific,reasonable,stable and feasible extraction method.【Method】Precise with yellow cake fall blood sugar in the active ingredient and index components flavonoids,total polysaccharide,glucose,ellagic acid,kaempferol-3-O-rutinosideand dry extract yield as evaluation index,with atmospheric circumfluence extraction L_(9)(3^(4))orthogonal experimental design method to optimize the extraction process parameters,the optimum processing parameters of atmospheric pressure extraction,the decompression by optimum parameters of atmospheric pressure extraction to extract,95%confidence interval overlapping method comparing the difference between the reduced pressure and atmospheric pressure from the component.【Result】Atmospheric pressure to extract the best technological parameters for:first use atmospheric pressure extraction time,alcohol and 12 times the amount of 70%ethanol,extract 60℃for 1 h,and then water,secondary water added to 12 times the amount of medicinal herbs,10 times,the water temperature of 90℃,2 h,1.5 h,decompression,amount of solvent extraction and alcohol concentration,extraction temperature,extraction time with normal pressure extraction,extraction pressure of 70%alcohol extraction only 150 hpa,the water 70 hpa,vacuum extracting ethanol-extraction,ellagic acid,total flavonoids content in liquid is higher than normal pressure extraction(P<0.05),vacuum extraction and water extraction liquid of glucose levels higher than normal pressure extraction(P<0.05).【Conclusion】The optimal process parameters of decompression extraction were 70%ethanol extraction 70 hPa and water extraction 150 hPa.First,70%ethanol extraction was extracted with decompression once,then 12 times of 70%ethanol was added for decompression extraction at 60℃for 1 h,and then extracted with water for two times.The amount of water added was 12 times and 10 times of the amount of medicinal materials,and the water extraction temperature was 90℃for 2h and 1.5 h.The decompression extraction technology is scientific,reasonable,stable and feasible,which can provide a basis for industrial production.
