姜黄素对APOE-/-动脉粥样硬化小鼠的保护作用及机制研究 被引量:5

摘要 目的探讨姜黄素对高脂诱导的动脉粥样硬化小鼠焦亡的影响。方法8周龄ApoE(-/-)小鼠30只随机分组。正常对照组6只;其余24只给予高脂饮食,其中单纯给予高脂饲料的6只小鼠归入空白对照组,另18只归入低浓度姜黄素组、中浓度姜黄素组、高浓度姜黄素组,每组6只。采集各组血清检测高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、甘油三酯(TG)、胆固醇(TC)、血糖(GLU)水平。制作各组小鼠颈动脉标本并行HE染色,通过酶标仪检测白细胞介素-1β(IL-1β)和白细胞介素-18(IL-18)水平;Western blot法检测Caspase-1水平。结果正常对照组血清HDL-C、LDL-C、TG、TC、GLU水平与空白对照组比较差异均有统计学意义(均P<0.05);高浓度姜黄素组血清上述指标水平与空白对照组、低浓度姜黄素组、中浓度姜黄素组比较差异均有统计学意义(均P<0.05)。正常对照组血清IL-1β、IL-18水平与空白对照组比较差异均有统计学意义(均P<0.05);高浓度姜黄素组血清上述指标水平与空白对照组、低浓度姜黄素组、中浓度姜黄素组比较差异均有统计学意义(均P<0.05)。高浓度姜黄素组Caspase-1水平明显低于空白对照组和低浓度姜黄素组,差异均有统计学意义(均P<0.05)。结论高浓度姜黄素延缓抗动脉粥样硬化进展,其作用机制可能与抗细胞焦亡有关。 Objective To investigate the effects of curcumin on atherosclerotic plaque and scorch death in mice with high lipid-induced atherosclerosis.Methods Thirty ApoE(-/-)mice aged 8 weeks were randomly divided into two groups.Normal control group 6;the other 24 mice were given high-fat diet,among which 6 mice given high-fat diet alone were assigned to blank control group,the other 18 mice were assigned to low-concentration curcumin group,medium-concentration curcumin group and high-concentration curcumin group,6 mice in each group.Serum levels of high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),triglyceride(TG),cholesterol(TC)and blood glucose(GLU)were detected in each group.Carotid artery samples of mice in each group were prepared and HE staining was performed to detect interleukin-1β(IL-1β)and interleukin-18(IL-18)levels by elisa.Caspase-1 levels were detected by Western Blot.Results The levels of serum HDL-C,LDL-C,TG,TC and GLU in normal control group were significantly different from those in blank control group(all P<0.05).There were statistically significant differences in serum levels of above indexes in high concentration curcumin group compared with blank control group,low concentration curcumin group and medium concentration curcumin group(all P<0.05).There were significant differences in serum IL-1βand IL-18 levels between normal control group and blank control group(all P<0.05).There were statistically significant differences in serum levels of above indexes in high concentration curcumin group compared with blank control group,low concentration curcumin group and medium concentration curcumin group(all P<0.05).Caspase-1 level in high curcumin group was significantly lower than that in blank control group and low curcumin group,the differences were statistically significant(P<0.05).Conclusion High concentration of curcumin delayed the progression of anti-atherosclerosis,and its mechanism may be related to anti-cell pyrophosis.
