摘要
目的探讨Na^(+)/K^(+)转运ATP酶α亚单位1(ATP1A1)在结肠腺癌(COAD)组织中的表达及其与COAD患者临床病理特征和预后的关系。方法实时荧光定量PCR检测ATP1A1在COAD组织和配对的癌旁组织中的表达,分析ATP1A1在COAD组织中的表达与患者临床病理特征和预后的相关性。结果与癌旁组织(5.70±5.87)相比,ATP1A1在COAD组织中的表达显著降低(2.96±3.98,t=4.966,P<0.001)。COAD组织中ATP1A1的表达与肿瘤大小(χ^(2)=5.537,P=0.019)、浸润深度(χ^(2)=5.326,P=0.021)和远处转移有关(χ^(2)=5.103,P=0.024)。ATP1A1低表达组COAD患者的中位生存时间(42.062个月)显著低于ATP1A1高表达组COAD患者(52.099个月,χ^(2)=9.954,P=0.002)。多因素COX回归模型分析表明,ATP1A1是COAD患者的独立预后因素,低表达ATP1A1预示着COAD患者的较差预后。结论ATP1A1在COAD组织中表达降低,与患者的预后密切相关,是COAD预后预测及治疗的潜在靶点。
Objective To investigate the expression of ATPase Na^(+)/K^(+) transporting subunit alpha 1(ATP1A1)in colonic adenocarcinoma(COAD),and to explore the relationship of ATP1A1 expression with clinicopathological characteristics and prognosis.Methods Quantitative real-time PCR was used to test the expression of ATP1A1 in the paired COAD and adjacent normal tissues.The associations of ATP1A1 expression with clinicopathological characteristics were evaluated.The prognostic value of ATP1A1 in the COAD patients were determined.Results The ATP1A1 expression was down-regulated in the COAD tissues compared with paired adjacent noncancerous tissues(2.96±3.98 and 5.70±5.87,t=4.966,P<0.001)and was related with tumor size(χ^(2)=5.537,P=0.019),T stage(χ^(2)=5.326,P=0.021)and distant metastasis(χ^(2)=5.103,P=0.024).The COAD patients with low ATP1A1 expression were associated with poorer overall survival compared to those with high ATP1A1 expression(42.062 months and 52.099 months,χ^(2)=9.954,P=0.002).COX analyses showed that low expression of ATP1A1 was an independent predictor for poor prognosis in COAD patients.Conclusion ATP1A1 expression was deceased in COAD,which presented a potential biomarker for poor prognosis of COAD patients and represented a possible molecular target for the treatment of COAD.
