摘要
目的研究新型光敏剂NHS-BODIPY-Br在体外对牙龈卟啉单胞菌(P gingivalis)和福赛坦菌(T forsythia)生物膜的抗菌作用。方法CCK-8检测光敏剂NHS-BODIPY-Br的生物相容性,将P gingivalis和T forsythia在厌氧(10%CO_(2),10%H_(2),80%N_(2))条件下混合培养48 h形成生物膜后进行抗菌光动力治疗,光敏剂浓度分别为5、10、15 mg/L。激光强度分别为15和30 J/cm^(2)。治疗后进行细菌活力检测、残留脂多糖检测以及激光共聚焦分析,并确定最佳治疗组合。结果当光敏剂NHS-BODIPY-Br的浓度为10 mg/L,光强度为15 J/cm^(2)时细菌生物膜减少率约98%。结论该研究说明用光敏剂NHS-BODIPY-Br进行抗菌光动力治疗可以有效地抑制P gingivalis和T forsythia生物膜。
Objective To study the antibacterial effect of a new photosensitizer NHS-BODIPY-Br on the biofilm of P gingivalis and T forsythia in vitro.Methods The biocompatibility of photosensitizer NHS-BODIPY-Br was detected by CCK8.P gingivalis and T forsythia were cultured in anaerobic condition(10%CO_(2),10%H_(2),80%N_(2))for 48 h to form a biofilm and then treated with antibacterial photodynamic therapy.The photosensitizer concentration was 5,10,15 mg/L and the laser intensity was 15 and 30 J/cm^(2).After treatment,bacterial activity,residual LPS and laser confocal analysis were performed to determine the optimal treatment combination.Results The reduction rate ofbacterial biofilm was about 98%when the concentration of NHS-BODIPY-Br was 10 mg/L and the laser intensity was 15 J/cm^(2).Conclusion This study shows that the NHS-BODIPY-Br-mediated photodynamic therapy can effectively inhibit the biofilm of P gingivalis and T forsythia.
