人参皂苷诱导宫颈癌HeLa细胞凋亡和自噬

摘要 目的探讨人参皂苷(Ginsenoside)影响宫颈癌HeLa细胞自噬与凋亡状况。方法本研究利用CCK-8法考察人参皂苷对细胞活力的作用;借助Annexin V/FITC-PI双染法检测人参皂苷对HeLa细胞凋亡情况;借助WB(Western blot)技术对凋亡相关蛋白的表达水平进行检测;利用AO染色检测自噬发生程度。结果 CCK-8法显示了人参皂苷Rh2和Rg3显著抑制了宫颈癌细胞的增殖,经细胞流式仪测定发现,对于细胞凋亡,Rh2与Rg3具促进作用,蛋白免疫印迹的结果表明Rh2和Rg3能够促进宫颈癌细胞凋亡因子的升高,同时吖啶橙染色的结果也表明人参皂苷Rh2和Rg3能够提高宫颈癌HeLa细胞自噬水平。结论人参皂苷Rh2和Rg3能够促进宫颈癌细胞凋亡并且调节其自噬。 Objective To explore the effect of ginsenosides on the apoptosis and autophagy of cervical cancer HeLa cells.Methods In this study,the CCK-8 method was used to investigate the effect of ginsenosides on cell viability;with the aid of Annexin V/FITC-PI double staining method,Ginsenoside affected the apoptosis of HeLa cells expand investigation;use WB(Western blot)technology to detect the expression level of apoptosis-related proteins;use AO staining to detect the degree of autophagy.Results The CCk-8 method found that ginsenosides Rh2 and Rg3 have a significant inhibitory effect on the proliferation of cervical cancer cells.The results of cell flow cytometry showed that Rh2 and Rg3 could promote cell apoptosis.The results of western blot showed that Rh2 and Rg3 could promote the increase of apoptotic factors in cervical cancer cells,and the results of acridine orange staining also indicate that ginsenosides Rh2 and Rg3 can increase the autophagy level of cervical cancer HeLa cells.Conclusion Ginsenosides Rh2 and Rg3 can promote the apoptosis of cervical cancer cells and regulate their autophagy.
