摘要
目的检测4种骨肉瘤细胞中Circ_0104346的表达,并通过下调该基因的表达,观察其对骨肉瘤细胞迁移及增殖的影响。方法通过收集4种骨肉瘤细胞,包括MG-63、U2OS、SaOS-2和SOSP-9607,荧光定量PCR测定细胞中的Circ_0104346的表达。选择表达量最高的一组骨肉瘤细胞,干扰Circ_010434的表达,观察其对骨肉瘤细胞迁移及增殖的影响。结果荧光定量PCR检测提示SaOS-2细胞系中的Circ_0104346的表达最高,选定该细胞系进行后续实验。干扰SaOS-2细胞Circ_010434的表达后,骨肉瘤细胞的迁移能力及增殖能力明显下降。结论骨肉瘤细胞中Circ_0104346表达上调,干扰Circ_0104346的表达能抑制其细胞迁移及增殖能力。
Objective Detected the expression of Circ_0104346 in four kinds of osteosarcoma cells,and observed its effect on the migration and proliferation of osteosarcoma cells by down-regulating the expression of this gene.Methods By collecting four types of osteosarcoma cells,including MG-63,U2OS,SaOS-2 and SOSP-9607,the expression of Circ_0104346 in the cells was determined by fluorescence quantitative PCR.Selected a group of osteosarcoma cells with the highest expression,interfere with the expression of Circ_010434,and observe its effect on the migration and proliferation of osteosarcoma cells.Results Fluorescence quantitative PCR detection indicated that the expression of Circ_0104346 in the SaOS-2 cell line was the highest,and this cell line was selected for subsequent experiments.After interfering with the expression of Circ_010434 in SaOS-2 cells,the migration and proliferation ability of osteosarcoma cells decreased significantly.Conclusion The expression of Circ_0104346 is up-regulated in osteosarcoma cells,and interference with the expression of Circ_0104346 can inhibit its cell migration and proliferation ability.
