摘要
MAPR蛋白家族在中枢神经系统大量表达,在神经细胞中具有神经营养作用。孕酮受体膜组分1(PGRMC1)、PGRMC2、神经营养因子蛋白(neudesin)和细胞色素b5结构域蛋白(neuferricin)都属于MAPR家族,是细胞色素b5家族的一个亚家族,由具有Cyt-b5样血红素/类固醇结合结构域的血红素结合蛋白组成。但与经典的核或膜结合类固醇受体缺乏同源性。除了结构上的相似性,PGRMC1、PGRMC2、neuferricin和neudesin表现出多功能的生理特性,涉及胆固醇/类固醇生物合成、药物代谢和反应、肿瘤发展、能量代谢、自噬、凋亡、细胞周期调节及神经功能。这篇综述简要总结了MAPR家族成员的结构和生理功能的研究现状。
MAPR protein family is highly expressed in the central nervous system and plays a neurotrophic role in nerve cells.Progesterone receptor membrane component 1(PGRMC1),PGRMC2,neurotrophin protein(neudesin)and cytochrome b5 domain protein(neuferricin)all belong to the MAPR family and are a subfamily of the cytochrome b5 family,which is composed of heme-binding proteins with Cyt-b5-like heme/steroid binding domains.However,it lacks homology with classical nuclear or membrane-bound steroid receptors.In addition to structural similarities,PGRMC1,PGRMC2,neuferricin and neudesin exhibit multifunctional physiological characteristics,involving cholesterol/steroid biosynthesis,drug metabolism and response,tumor development,energy metabolism,autophagy,apoptosis,cell cycle regulation,and neurological function.This review briefly summarizes the research status of the structure and physiological function of MAPR family members.
