摘要
为探讨梅公散的抗炎作用及其作用机制,开展梅公散对炎症模型小鼠血液和组织中前列腺素E_(2)(PGE_(2))含量的研究。试验采用二甲苯致炎法和小鼠足跖肿胀法研究梅公散对小鼠急性炎症的抗炎作用;采用竞争性ELISA法测定小鼠血清中和肿胀足中PGE_(2)的含量。结果表明,梅公散可显著降低小鼠耳廓肿胀度(P<0.05);致炎4 h后,可极显著抑制蛋清所致小鼠足趾肿胀(P<0.01);致炎2 h后,低剂量的梅公散可极显著抑制甲醛所致小鼠足趾肿胀(P<0.01),高剂量的梅公散可显著抑制甲醛所致小鼠足趾肿胀(P<0.05)。高剂量的梅公散可显著降低二甲苯所致小鼠血清中PGE2含量(P<0.05),低剂量的梅公散则对血清中PGE2含量则无显著作用(P>0.05);高剂量的梅公散可显著降低甲醛和蛋清所致小鼠肿胀足中PGE_(2)的含量(P<0.05)。研究表明,梅公散对小鼠血液和组织中PGE_(2)含量的影响与使用剂量存在相关性,且梅公散抗炎作用的产生机制与降低炎症模型小鼠血液和组织中PGE2的含量有关。
In order to clarify the anti-inflammatory effect and its mechanism of Meigong powder,the effect of Meigong powder and its influence on PGE_(2) content in blood and tissues of inflammatory model mice.The anti-inflammatory effect of Meigong powder on acute inflammation in mice were studied by the methods of xylene-induced inflammation and paw swelling in mice,the contents of PGE_(2) in serum and swollen feet of mice were determined by the method of competitive ELISA.The results showed that Meigong powder could significantly reduce the swelling of the ears of mice(P<0.05).Four hours after the inflammation,it could significantly inhibite the toe swelling of mice caused by egg white(P<0.01).Two hours after the inflammation,low-dose Meigong powder could significantly inhibite the toe swelling of mice caused by formaldehyde(P<0.01),high-dose Meigong powder could significantly inhibite the toe swelling of mice caused by formaldehyde(P<0.05).High-dose Meigong powder could significantly reduce the PGE_(2) content in the serum of mice caused by xylene(P<0.05),while low-dose Meigong powder had no significant effect on the serum PGE_(2) content(P>0.05).High-dose Meigong powder could significantly reduce the content of PGE_(2) in mice swollen feet caused by formaldehyde and egg white(P<0.05).Studies have shown that the effect of Meigong powder on the PGE_(2) content in the blood and tissues of mice is related to the dosage,and the mechanism of the anti-inflammatory effect of Meigong powder is related to the reduction of the PGE_(2) content in the blood and tissues of the inflammation model mice.
