Effects of FSP27 Gene on Proliferation,Differentiation and Apoptosis of Porcine Adipocytes

摘要 为研究FSP27基因对猪前体脂肪细胞增殖、分化和凋亡的影响,在军牧一号白猪原代前体脂肪细胞中转染siRNA-717沉默FSP27。猪前体脂肪细胞分化效果使用油红O染色试验检测,猪前体脂肪细胞增殖、分化、凋亡相关基因的表达使用荧光定量PCR(RT-qPCR)、蛋白免疫印迹等技术检测。结果表明:猪脂肪细胞中转染siRNA-FSP27后,PCNA,cyclin D2和cyclin E2的mRNA表达水平显著升高;aP2、C/EBPα、PPARγ的mRNA表达水平显著降低;油红O检测结果显示脂滴显著降低;Bax和p53的相对mRNA水平显著降低,Bcl2显著升高。表明FSP27在体外培养的猪脂肪细胞中抑制脂肪细胞增殖,促进脂肪细胞分化和凋亡。 In order to study the effects of FSP27 on proliferation,differentiation and apoptosis of porcine preadipocytes,siRNA-717 was transfected into the primary preadipocytes of pig Junmu No.1 white pig to silence FSP27.Oil red O staining was used to detect the differentiation effect of preadipocytes.Real-time PCR(RT-qPCR) and Western blotting were used to detect changes in proliferation,differentiation,and apoptosis-related gene expression in pig preadipocytes.The results showed that after transfecting siRNA,the mRNA expression levels of PCNA,cyclin D2 and cyclin E2 were significantly increased in pig preadipocytes;the mRNA expression levels of aP2,C/EBPα and PPARγ were significantly decreased;oil red O test results showed lipid droplets were significantly decreased;the relative mRNA levels of Bax and p53 were significantly decreased,but Bcl2 was significantly increased.This indicates that FSP27 can inhibit the proliferation of preadipocytes,and promote the differentiation and apoptosis of preadipocytes in porcine preadipocytes cultured in vitro.
