Analysis of Bacterial Community Structure and Diversity in Corn Steep Liquor By-Products Based on High Throughput Sequencing Technology

摘要 为了更加全面了解玉米浸泡副产物中微生物多样性,比较其中细菌群落结构,利用高通量测序技术,对玉米浸泡液、玉米浆及玉米喷浆粉中细菌16S rRNA V4区进行测序,进而分析细菌多样性和群落组成结构。结果表明:共获得190069条有效数据,通过聚类获得352个OTUs,隶属于12门、68纲、183属。α多样性分析结果表明,玉米喷浆粉中Shannon指数明显高于玉米浸泡液和玉米浆。微生物群落组成分析发现,在门水平上,厚壁菌门(Firmicutes)是玉米浸泡液、玉米浆及玉米喷浆粉的共同优势菌门;在属水平上,乳杆菌属(Lactobacillus)为玉米浸泡液、玉米浆及玉米喷浆粉的优势属;在种水平上,玉米浸泡液中优势菌为耐酸乳杆菌(L.acetotolerans),玉米浆及玉米喷浆粉中优势菌为解淀粉乳杆菌(L.amylolyticus)。 In order to fully demonstrate the microbial diversity in corn steep liquor by-products and to compare the community composition,diversity and community structure of bacterial in corn steepwater,corn steep liquor and dried corn steep liquor powder were analyzed based on high throughput sequencing the V4 region of 16 S rRNA.The results showed that a total of 190069 effective tags were obtained,and 352 OTUs were obtained by cluster analysis,which belonged to 12 phyla,68 classes and 183 genera.Alpha diversity analysis showed that the Shannon-Wiener diversity index of dried corn steep liquor powder was higher than that of corn steepwater and corn steep liquor.At the phyla level,Firmicutes was the predominant phylum of corn steepwater,corn steep liquor and dried corn steep liquor powder.At the genera level,Lactobacillus was dominant.At the species level,L.acetotolerans was the most abundant bacteria in corn steepwater,L.amylolyticus was the major one in corn steep liquor and dried corn steep liquor powder.
