miR-203a-3p对胰腺癌细胞系Panc-1生长增殖、迁移侵袭的促进作用 被引量:1

摘要 目的研究微小RNA-203a-3p (miR-203a-3p)对胰腺癌细胞系Panc-1增殖、集落形成以及迁移侵袭的影响,确定其潜在机制,为胰腺癌患者的临床诊治提供参考。方法通过实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测miR-203a-3p在胰腺癌细胞株Panc-1、Aspc-1以及人胰腺导管上皮细胞(HPNE)中的表达水平;将miR-203a-3p模拟物(miR-203a-3p mimics)、miR-203a-3p抑制物(miR-203a-3p inhibitor)及其阴性对照NC mimics、NC inhibitor转染到Panc-1中。实验组分为NC mimics组、miR-203a-3p mimics组、NC inhibitor组、miR-203a-3p inhibitor组。CCK-8实验和集落形成实验分别检测细胞增殖活性和细胞集落形成数量,Transwell实验检测迁移细胞和侵袭细胞数量。结果 qRT-PCR实验结果显示在Panc-1、Aspc-1细胞中,miR-203a-3p的表达水平分别上升了7.71倍和6.15倍,差异具有统计学意义(P<0.05);过表达miR-203a-3p后Panc-1细胞系中miR-203a-3p表达水平升高了27.94倍,抑制miR-203a-3p表达后Panc-1细胞系中miR-203a-3p表达水平降低了0.47倍,与相应对照组比较差异具有统计学意义(P<0.05);过表达miR-203a-3p后促进了Panc-1的增殖、集落形成和迁移侵袭;细胞中转染miR-203a-3p inhibitor后,抑制了Panc-1的增殖、集落形成,迁移侵袭能力受到抑制(P<0.05)。结论 miR-203a-3p能够促进胰腺癌细胞系Panc-1生长增殖及迁移侵袭能力。 Objective To study the effection of miR-203a-3p on the cell proliferation ability, colony-forming ability, migration ability and invasion ability of pancreatic cancer cell line Panc-1, so as to provide reference for clinical treatment of pancreatic cancer.Methods The expression levels of miR-203a-3p in pancreatic cancer cell lines Panc-1, Aspc-1 and human normal pancreatic epithelial cells HPNE were detected by qRT-PCR. The experiment was divided into NC mimics group, miR-203a-3p mimics group, NC inhibitor group and miR-203a-3p inhibitor group. CCK-8 and colony-forming assays were used to detect cell proliferation activity and colony formation number, respectively. Transwell assay was used to detect the number of migrating cells and invasive cells.Results The results of qRT-PCR showed that the expression levels of miR-203a-3p in Panc-1 and Aspc-1 were increased by 7.71 and 6.15 times, respectively, and the difference was statistically significant(P<0.05). The result of qRT-PCR showed that the miR-203a-3p level was increased by 27.94 times after transfection of miR-203a-3p mimics compared with NC mimics group and the expression level of miR-203a-3p was decreased by 0.47 times after transfection of miR-203a-3p inhibitor compared with NC inhibitor group, and the difference was statistically significant(P<0.05). miR-203a-3p mimics promoted the proliferation, colony-forming, migration ability and invasion ability of the Panc-1. After transfection of miR-203a-3p inhibitor into Panc-1, the cell proliferation, cell colony-forming, cell migration ability and invasion ability of Panc-1 were inhibited(P<0.05).Conclusion miR-203a-3p may promote the proliferation, migration and invasion ability of the Panc-1.
