中药注射剂喜炎平对阿奇霉素组织分布的影响

摘要 本研究旨在探讨中药注射剂喜炎平(XYP)对阿奇霉素(AZM)在肺炎模型大鼠体内组织分布的影响。将肺炎模型大鼠随机分为对照组和实验组,分别经尾静脉注射AZM和/或XYP,剂量均为63.0 mg/kg,每日1次,连续给药1周。LC-MS/MS法测定末次给药24 h后大鼠血浆和组织中AZM浓度,并比较对照组与实验组AZM的组织分布差异。结果显示,建立的LC-MS/MS方法经考察符合生物样品分析要求,适用于AZM浓度测定。与对照组相比,实验组AZM在血浆、心、脾、肺和肾组织中没有显著性差异,在肝脏中显著降低(P<0.05)。研究结果表明XYP对AZM在肝脏的分布有显著影响,降低其在肝中的药物蓄积,可能与XYP促进AZM代谢有关,有利于减轻长期使用AZM造成的肝毒性。 The purpose of this study is to investigate the effect of Chinese medicine injection Xiyanping(XYP)on the tissue distribution of azithromycin(AZM)in rats with pneumonia.The pneumonia model rats are randomly divided into a control group and an experimental group.AZM and/or XYP are injected via the tail vein at a dose of 63.0 mg/kg,once a day for one week.The LC-MS/MS method is used to determine the concentration of AZM in rat plasma and tissue 24 h after the last administration,and to compare the tissue distribution of AZM between the control group and the experimental group.The results show that the established LC-MS/MS method meets the requirements of biological sample analysis and is suitable for the determination of AZM concentration.Compared with the control group,AZM in the experimental group has no significant differences in plasma,heart,spleen,lung,and kidney tissues,but significantly decreases in the liver(P<0.05).It is concluded that XYP has an obvious impact on the distribution of AZM in the liver,which may be because XIP can promot the metabolism of AZM and thus reduce drug accumulation,and is beneficial to alleviate the liver toxicity caused by long-term use of AZM.
