中药复方和益生素对热应激大鼠肠道菌群的影响

摘要 本试验旨在探讨大鼠热应激与肠道菌群变化的关系,并为中药复方和益生素干预热应激的作用机制提供依据。大鼠热应激下灌服中药复方和等体积益生素后于热应激3 h、1 d、3 d、7 d后,检测大鼠血清HSP70蛋白浓度;收集盲肠段内容物,提取总DNA进行16S rDNA测序,分析肠道微生物组分和多样性。结果显示:热应激各时段热应激组HSP70蛋白浓度极显著升高(P<0.01),中药组和益生素组肠道菌群多样性比热应激组显著增加(P<0.05)。热应激下肠道优势菌属厚壁菌和拟杆菌相对减少(P>0.05),乳酸菌属极显著减少(P<0.01),中药组的拟杆菌属和益生素组的乳酸菌属与热应激组相比显著升高(P<0.05)。京都基因和基因组数据库(KEGG)统计分析可知,热应激组在糖代谢、氨基酸代谢和膜转运的基因表达活跃,中药组、益生素组和常温空白组KEGG途径相似,中药组生物路径上基因活动最低,益生素组KEGG途径与常温空白组接近。 The aim of this study was to investigate the relationship between heat stress and intestinal flora changes in rats,and to provide experimental evidence for the action mechanism of Chinese herbal compound and probiotics in relieving heat stress.The rats were fed with traditional Chinese medicine and equal volume probiotics under heat stress and their serum HSP70 protein concentration was determined at 3 h,1 d,3 d and 7 d after heat stress.The contents of cecal segment were collected,and the total DNA was extracted for 16S rDNA sequencing to analyze the intestinal microbial composition and diversity.The results showed that the concentration of HSP70 protein in heat stress group increased significantly at different time points(P<0.01).The diversity of intestinal flora in Chinese medicine group and probiotics group was significantly higher than that in heat stress group(P<0.05).In heat stress group,the dominant bacteria in the intestinal tract were significantly decreased(P>0.05),and Lactobacillus was significantly decreased(P<0.01).Compared with the heat stress group,Bacteroides in traditional Chinese medicine group and Lactobacillus in probiotics group were significantly increased(P<0.05).Statistical analysis of functional gene KEGG revealed that the gene expression of glucose metabolism,amino acid metabolism and membrane transport were active in heat stress group.The KEGG pathway of traditional Chinese medicine group,probiotics group and control group was similar,with gene activity in biological pathway of traditional Chinese medicine group being the lowest,and KEGG pathway of probiotics group was close to that of control group.
