脊尾白虾对低温胁迫响应的转录组学分析

摘要 温度是影响甲壳类动物多种生命活动包括生长发育、生理代谢、繁殖及分布等的重要因子。以脊尾白虾(Exopalaemon carinicauda)为研究对象,分析其耐低温生存的分子机制。采用Illumina HiSeq 2500高通量测序平台,对低温组(0℃)和对照组(25℃)脊尾白虾整虾分别进行转录组测序,测序数据质控、拼接和组装后获得69997个Unigenes,经BLAST软件与NR,Swiss-Prot,GO,COG和KEGG等数据库比对注释,最终获得注释Unigenes 23098个。差异表达基因(differentially expressed gene,DEG)GO富集分析表明,568条DEG被注释到胞外区(Extracellular region)、结构分子活性(Structural molecule activity)、细胞膜(Cytomembrane)、细胞连接(Cell junction)、分子传感器活性(Molecular transducer activity)、信号传导(Signal transduction)和生物附着(Biological adhesion)等GO条目;KEGG通路分析发现部分DEG在核糖体(Ribosomal)、霍乱弧菌感染(Vibrio cholerae infection)、氨基糖和核苷酸糖代谢(Amino sugar and nucleotide sugar metabolism)等信号通路中富集。该研究丰富了对脊尾白虾基因资源的认识,为进一步探究其耐低温机制提供了基础数据。 Temperature,as an important factor,affects a variety of life activities of crustaceans,including its growth and development,physiological metabolism,reproduction and distribution.In order to analyze the molecular mechanism of low-temperature survival,E.carinicauda in the low-temperature(0℃)and those in the control group(25℃)were sequenced by Illumina HiSeq 2500 high-throughput sequencing platform.69997 Unigenes were obtained after sequencing data quality control,splicing and assembly.After BLAST software and NR,Swiss-Prot,GO,COG and KEGG databases compared noting,23098 Unigenes were finally obtained.GO enrichment analysis of differentially expressed gene(DEG)showed that 568 DEGs were annotated to GO items such as Extracellular region,Structural molecular activity,Cytomembrane,Cell junction,Molecular transducer activity,Signal transduction and Biological adhesion;KEGG pathway analysis found that some DEG was enriched in Ribosomal,Vibrio cholerae infection,Amino sugar and nucleotide sugar metabolism.This study enriches the gene resources of E.carinicauda,and provides basic data for further exploring its low temperature tolerance mechanism.
