养殖红鳍东方鲀的肠和胃转录组比较分析

摘要 为了探究养殖红鳍东方鲀肠组织和胃组织在主要功能上的差异性,本研究选择一龄红鳍东方鲀通过转录组测序方法,比较肠和胃2种组织在基因表达上的差异性。研究分析发现:在胃组织中测出的碱基数均大于4.89×10^(7) bp,在肠组织中测出的碱基数均大于5.42×10^(7) bp。在肠相对胃(SA13 vs IA13)的比较中共获得3564个差异表达基因,其中2167个上调表达,1397个下调表达。在GO富集结果中的酶调节活动、细胞骨架结合、跨膜转运蛋白活性等功能存在显著富集。在KEGG的富集分析中发现大量与代谢有关的通路存在显著富集,这些代谢通路中大多数的差异基因在肠中表达量高于胃,呈下调表达。此外在粘着连接、紧密连接、细胞粘附分子等通路中也存在显著富集,而其中大多数的基因呈上调表达。研究中筛选到6个差异表达基因并通过RT-qPCR方法进行验证,验证结果与转录组测序结果保持一致。研究结果为今后养殖红鳍东方鲀饵料选择和提高饵料系数提供思路,也为其他生物肠道基因的研究提供参考。 In order to investigate the differences in the main functions between the intestine and stomach of farmed Takifugu rubripes,this study selected one-year-old T.rubripes to compare the differences in gene expression between intestinal and stomach tissues by transcriptome sequencing.The bases measured in intestinal and stomach tissues were greater than 4.89×10^(7) bp and 5.42×10^(7) bp,respectively.A total of 3564 differentially expressed genes were obtained from the comparison of intestine versus stomach(SA13 vs IA13),among which 2167 genes were up-regulated and 1397 genes were down-regulated.There was significant enrichment of enzyme regulatory activity,cytoskeletal protein binding,and active transmembrane transporter activity in the GO enrichment results.The KEGG enrichment analysis revealed significant enrichment in a large number of metabolism-related pathways,and most of the differentially expressed bases in these metabolic pathways were down-regulated in the intestine compared to the stomach.In addition,there were significant enrichments in the adhesion,tight junctions and cell adhesion molecules pathways,and most of the genes were up-regulated.Six differentially expressed genes were screened and validated by RT-qPCR method,and the validation results were consistent with the transcriptome sequencing results.The results of this study will provide ideas for future bait selection and improvement of bait coefficient in the culture of T.rubripes and provide reference for the study of intestinal genes in other species.
