中俄边境黑瞎子岛鼠群中巴尔通体调查 被引量:1

Survey on Bartonella in rodents collected at Heixiazi Island of Sino-Russian border

摘要 目的明确中俄边境黑瞎子岛鼠群中巴尔通体(Bartonella)的分布和分子流行特征。方法2017年4-10月,在黑瞎子岛用夹夜法捕鼠,解剖取鼠肝和脾脏样本,用兔血脑心浸液琼脂培养基分离培养巴尔通体,用PCR法扩增巴尔通体柠檬酸合成基因(gltA)片段,阳性者经基因测序,进行系统发育分析。结果共捕鼠354只,分属于3科6属6种。鼠类巴尔通体感染率为49.72%,所有鼠种均有感染,红背鼠平感染率最高(57.97%),其次为东方田鼠(53.85%),黑线姬鼠(41.41%)和大林姬鼠(40.00%)感染率也较高,褐家鼠(33.33%)和花鼠(20.00%)也有感染。共分离到18株巴尔通体,系统进化分析显示鼠类携带的巴尔通体分属于5个种群:泰勒巴尔通体(Bartonella taylorii)、格拉范姆巴尔通体(Bartonella grahamii)、Bartonella rudakovii、文森巴尔通体(Bartonella vinsonii)、道志巴尔通体(B.doshiae)。优势鼠种红背鼠平感染了4种巴尔通体(B.taylorii、B.grahamii、B.vinsonii、B.doshiae),而第二优势鼠种黑线姬鼠仅感染1种巴尔通体(B.grahamii);东方田鼠感染4种巴尔通体(B.taylorii、B.grahamii、B.vinsonii、B.rudakovii)。结论中俄边境黑瞎子岛的鼠类普遍感染巴尔通体,且存在种类和宿主的多样性和特异性。 Objective To know the distribution and molecular characteristics of Bartonella on the port area of Heixiazi Island between China and Russia.Methods The rodents were collected by night-clamping on the island,April to Octomber,2017.The spleen and liver samples were got with aseptic operation.Bartonella was isolated on Brain Heart Infusion Agar Media supplemented with Rabbit Blood.PCR was used to amplified Bartonella citrate synthase(glt A)gene,then the positive fragment was sequenced and phylogenetic analyzed.Results A total of 354 rodents were captured,belonging to 3 families,6 genera 6 specises.The Bartonella infection rate was 49.72%in rodent groups.The prevalence of Bartonella bacteremia existed in all the species of rodents,with the highest infection rate of Clethrionomys rutilus(57.97%),followed by Microtus fortis Buchner(53.85%),Apodemus agrarius(41.41%),Apodemus peninsulae(40.00%),Rattus norvegicus(33.33%)and Eutamias sibiricus(20.00%).Totally 18 Bartonella strains were isolated,the phylogenetic analysis showed that Bartonella strains were belonged to five branches including B.taylorii,B.grahamii,B.rudakovii,B.vinsonii,B.doshiae.The dominant species of the rodent,Clethrionomys rutilus,was found infected with four species of Bartonella,such as B.taylorii,B.grahamii,B.vinsonii and B.doshiae.While the second major species of rodent hosts,Apodemus agrarius only infected with B.grahamii.The host Microtus fortis infected with four species of Bartonella,such as B.taylorii,B.grahamii,B.vinsonii and B.rudakovii.Conclusion It is confirmed that rodents on Heixiazi Island were widely infected with different Bartonella,the hosts and Bartonella were varied and specified as well.
