摘要
通过双相酸水解法优化山奈酚提取条件,结合高效液相色谱(HPLC)法,通过单因素实验考察酸浓度、提取时间和固液比对山奈酚提取率的影响,并利用响应面Box-Behnken实验优化提取山奈酚的最佳工艺.实验结果表明:最佳提取条件的盐酸浓度为6.1×10^(-2) mmol/L,提取时间为12.87 min,固液比为1∶137.46,提取率达1.11%;山奈酚具有一定的抗氧化作用,对超氧自由基(·O^(2-))和1,1-二苯基-2-三硝基苯肼(DPPH·)均有一定的清除能力,尤其对DPPH·的清除能力更强,且清除能力随浓度的升高而提高.
The extraction conditions of kaempferol were optimized by a two-phase acid hydrolysis method,and the effects of acid concentration,extraction time and solid-liquid ratio on the extraction yield of kaempferol were investigated by a single factor experiment combined with high performance liquid chromatography(HPLC).The response surface Box-Behnken experiment was used to optimize the extraction process of kaempferol.The experimental results show that the optimum extraction conditions are the concentration of hydrochloric acid of 6.1×10^(-2) mmol/L,the extraction time of 12.87 min,the solid-liquid ratio of 1∶137.46,and the extraction yield is 1.11%.Kaempferol has a certain antioxidant effect,and has certain scavenging ability on superoxide free radical(·O^(2-))and 1,1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl(DPPH·),especially on DPPH·,and the scavenging ability increases with the increase of concentration.
