建立兔VX2肝癌模型及血管介入操作方法

摘要 目的:总结建立兔VX2肝癌模型和血管介入操作的经验和要点。方法:建立兔VX2肝癌模型70只,随机挑选60只分为三组,并在术后7天、14天、21天分别选定一组行MRI检查和肝动脉及门静脉DSA造影,分析肿瘤大小、影像学特征和肺转移情况。结果:建模成功69只(成功率98.6%)。成瘤后第14天MRI检查显示肿瘤呈较长T1、长T2信号,增强扫描肿瘤呈环形强化,DWI成像呈显著高信号,术后第7天、14天和21天,肿瘤最大直径分别为(1.11±0.36)cm、(2.32±0.50)cm和(3.56±0.67)cm,发生肺转移率为10%、20%和80%,随着时间延长肿瘤生长速度加快(P<0.01)且肺转移发生率升高(P<0.05)。DSA显示随时间延长富血供肿瘤发生率明显增加,分别为25%、80%和95%。结论:兔VX2肝癌模型建模成功率高,血管内介入操作可行性强,综合考虑植瘤后14天左右为最佳介入操作时间窗。 Objective:To summarize the experience and key points of establishing rabbit VX2 liver carcinoma model and endovascular interventional procedure.Methods:70 New Zealand white rabbits underwent laparotomy were used to establish the VX2 liver carcinoma model.60 rabbits were randomly divided into three groups.One group was selected to undergo MRI examination and hepatic artery and portal vein DSA on the 7th,14th and 21st day after operation,respectively,to analyze the changes in tumor size,imaging characteristics and pulmonary metastasis.Results:69 out of 70 experimental rabbits were successfully modeled(98.6%).MRI examination on the 14th day after tumor formation showed that the tumor presented long T1 and long T2 signals,the tumor showed circular enhancement on enhanced scan,and the tumor tissue on DWI showed high signal.Respectively,the maximum diameter of the tumor was(1.11±0.36)cm,(2.32±0.50)cm and(3.56±0.67)cm,and the incidence of lung metastasis was 10%,20%and 80%on the 7th day,14th day and 21st day after operation.Apparently,the tumor growth rate(P<0.01)and the incidence of lung metastasis increased significantly with time(P<0.05).DSA showed that the incidence of blood-rich tumors increased significantly with time,which were 25%,80%,and 95%.Conclusion:The rabbit VX2 liver cancer model is an excellent animal model used in interventional studies.It can be successfully modeled easily,and has strong feasibility in endovascular interventional procedure.The 14th day or so after tumor implantation is the best time for endovascular intervention.
