摘要 以金菠萝吸芽或腋芽为外植体,探讨不同消毒方法、不同生长调节剂配比对芽诱导、继代增殖和生根培养的影响。结果表明,用75%酒精消毒30 s再用0.1%HgCl_(2)浸泡20~25 min效果最好;适宜菠萝吸芽诱导和继代增殖培养的培养基为MS+6-BA 3.0 mg/L+NAA 0.1 mg/L,增殖系数为3.19,继代增殖周期为25~30 d较为适宜;适宜生根培养基为1/2 MS+NAA 1.5 mg/L,其生根率达到100%。该研究为完善金菠萝组培快繁技术提供依据。 Suckers and axillary buds of Golden Pineapple were used as explants for tissue culture.The effects of different sterilization methods and medium components on the induction of shoots,subculture for multiplicaton and rooting culture were studied. The results showed that sterilization with 0.1% HgCl2 for 20-25 min after sterilization with 75% alcohol for 30 s had the best effect on the explants and that the optimal medium for shoot induction and subculture for multiplication is MS+6-BA(3.0 mg·L^(-1)) +NAA(0.1 mg·L^(-1)), which has a multiplication coefficient of 3.19 and a subculture cycle of 25 to 30 days. The optimal medium for rooting is 1/2 MS+NAA(1.5 mg·L^(-1)) with the rooting rate of 100%. This study provides some references for improving the fast propagation of Golden Pineapple via tissue culture.
