摘要
目的观察沙利度胺(THD)对人宫颈癌Hela细胞周期形成的影响。方法利用不同浓度的THD(50μg/mL、100μg/mL与200μg/mL)处理人宫颈癌Hela细胞,利用流式细胞术检测细胞周期分布情况。结果不同浓度的THD作用于人宫颈癌Hela细胞,能明显降低细胞活力,且体现出浓度依赖性(P<0.05);作用24 h后,Hela细胞G0/G1期细胞数目增加显著(P<0.05);S期细胞显著减少(P<0.05)。另外,THD持续处理Hela细胞24 h后,p-p53/p53比值显著增加(P<0.05)。结论THD对人宫颈癌Hela细胞周期形成有较明显的抑制作用。
Objective To observe the effect of thalidomide(THD)on the cell cycle formation of Hela in human cervical cancer.Methods Human cervical cancer Hela cells were treated with different concentrations of THD(50μg/mL,100μg/mL and 200μg/mL),and the cell cycle distribution was detected by flow cytometry.Results THD at different concentrations acted on Hela cells of human cervical cancer,which significantly reduced cell activity and showed concentration dependence(P<0.05).After 24 hours,the number of cells in Hela cells G_(0)/G_(1) phase increased significantly(P<0.05);S-phase cells were significantly reduced(P<0.05).In addition,after THD continued to treat Hela cells at 24 hours,the p-p53/p53 ratio increased significantly(P<0.05).Conclusion THD can significantly inhibit the cell cycle of Hela in human cervical cancer.
