摘要
铁死亡是一种铁依赖性的由脂质过氧化引起的新型细胞死亡方式,区别于细胞凋亡、坏死、焦亡及自噬等,其主要特征是铁蓄积和脂质过氧化。铁死亡与氨基酸、脂质和铁代谢密切相关,并对癌症、衰老、神经退行性疾病、缺血再灌注损伤等疾病有重要影响。本文主要综述了铁死亡的分子机制以及靶向铁死亡的相关疾病的治疗策略,并对其未来研究可能存在的问题与方向进行了分析和展望。
Ferroptosis is a new type of cell death caused by iron dependent lipid peroxidation,which is different from apoptosis,necrosis,proptosis and autophagy.It characterized by the iron accumulation and lipid peroxidation.Ferroptosis is regulated by the metabolism of amino acids,lipids and iron,and plays a crucial role in cancer,aging,neurodegenerative diseases and ischemia reperfusion injury.Here,we will focus on the recent advances in understanding the regulatory mechanisms of ferroptosis,and summarize the development of the therapeutic strategies that target ferroptosis,and the problems and directions of its future research.In addition,the certain problems and research directions arising in the field of ferroptosis are also analyzed and presented.
