油桐花、叶、果壳代谢组成分鉴定与差异分析 被引量:3

Identification and differential analysis of metabolomic composition of leaves, flowers and shells of Vernicia fordii

摘要 油桐(Vernicia fordii)是一种优势明显的生物柴油生产原料。针对油桐各组织中的代谢产物的利用开发仍处在初级阶段,因此探究油桐不同组织的代谢产物具有重要的实际意义。本文利用超高效液相色谱-质谱联用(UPLC-MS/MS)的广泛靶向代谢组学技术对油桐花、叶及果壳中代谢产物进行鉴定,并对花和叶中的差异代谢物进行分析。结果表明:花、叶、果壳三个组织中共鉴定出944种代谢产物,其中共有代谢产物为462种;油桐果壳中的特异代谢产物有348种,以酚类、黄酮和生物碱类居多,而花和叶中的特异代谢产物分别为9种和2种,以萜类、黄酮和香豆素类为主,果壳中的特有代谢产物显著多于花与叶;油桐花和叶的差异代谢物有596种,主要分布在69条代谢途径中,其中富集较多的5条通路分别为嘌呤代谢通路、苯丙烷类代谢合成通路、环戊酮-2-羧酸甲酯代谢通路、氨酰-tRNA生物合成通路,以及苯丙氨酸、酪氨酸和色氨酸生物合成通路。本研究揭示了油桐花、叶、果壳中的特异代谢产物,为油桐副产品定向开发利用提供科学依据。 Vernicia fordii is an advantageous raw material for biodiesel production. The exploitation of the metabolites in the tissues of V. fordii is still at an early stage, so it is of great practical importance to investigate the metabolites in the different tissues of V. fordii. In this paper, extensive targeting metabolomic determination method with ultra-performance liquid chromatography–tandem mass spectrometry(UPLC-MS/MS) was used for the identification of metabolites in flowers, leaves and shells of V. fordii and analysis of differential metabolites in flowers and leaves. The results show that a total of 944 metabolites were identified in the three tissues, of which 462 were common metabolites. There are 348 specific metabolites in shells of V. fordii, the majority of which are phenols, flavonoids and alkaloids. There are 9 and 2 specific metabolites in flowers and leaves of V. fordii, respectively, the majority of which are terpenoids, flavonoids and coumarins. Specific metabolites in shells are significantly more than those in flowers and leaves.There are 596 differential metabolites of V. fordii flowers and leaves, mainly distributed in 69 metabolic pathways, of which five pathways with high enrichment are purine metabolism, phenylpropanoid biosynthesis, 2-oxocarboxylic acid metabolism, aminoacyl-tRNA biosynthesis and phenylalanine, tyrosine and tryptophan biosynthesis. This study reveals the specific metabolites in the flowers, leaves and shells of V.fordii, providing a scientific basis for the targeted exploitation of V. fordii by-products.
