蜻蜓石斛类原球茎的诱导与植株再生系统建立 被引量:2

Induction of protocorm-like bodies and establishment of plant regeneration system in Dendrobium pulchellum

摘要 以蜻蜓石斛(Dendrobium pulchellum)茎尖为外植体,研究了不同植物生长调节剂、天然有机添加物对类原球茎(protocorm-like bodies, PLBs)诱导以及丛生芽增殖及生长的影响。结果表明:最适合PLBs诱导和分化的培养基为MS+2.5 mg·L^(-1)TDZ+0.1 mg·L^(-1)NAA;最佳丛生芽增殖培养基为MS+0.8 mg·L^(-1)6-BA+0.5 mg·L^(-1)NAA;有机添加物香蕉泥可以有效促进丛生芽苗高和根系生长,而桃子泥、苹果泥有利于提高丛生芽的增殖效率。 To achieve the mass-scale multiplication of Dendrobium pulchellum, the optimal media for induction and differentiation of protocorm-like bodies(PLBs), multiplication and growth for shoots were studied and the rapid propagation system by tissue culture was established. The results showed that MS + 2.5 mg·L^(-1)TDZ + 0.1 mg·L^(-1)NAA was optimal for PLBs induction and differentiation. MS + 0.8 mg·L^(-1)6-BA +0.5 mg·L^(-1)NAA was the most suitable for multiplication for shoots. The organic amendment banana homogenate could effectively promote the plant height and root growth, while peach and apple homogenate were helpful to improve the multiplication efficiency of shoots.
