长链非编码RNA PSMA3-AS1在非小细胞肺癌中的表达及其对细胞侵袭和凋亡的影响 被引量:1

Expression of long non-coding RNA PSMA3-AS1 in non-small cell lung cancer and its effect on cell invasion and apoptosis

摘要 目的探讨长链非编码(lncRNA)PSMA3-AS1在非小细胞肺癌(NSCLC)组织中的表达以及其对NSCLC细胞H1299侵袭和凋亡的影响。方法选取NSCLC组织及与其配对的癌旁正常组织(作为对照)各50例,用实时荧光定量聚合酶链反应(RT-qPCR)法检测PSMA3-AS1在NSCLC组织和癌旁正常组织中的相对表达水平;将H1299细胞分为对照组、实验1组、实验2组,对照组转染si-NC,实验1组转染si-PSMA3-AS1-1,实验2组转染si-PSMA3-AS1-2。用噻唑蓝(MTT)实验检测细胞增殖水平,用Transwell实验检测细胞侵袭水平,用流式细胞仪检测细胞凋亡水平。结果NSCLC组织与癌旁正常组织中PSMA3-AS1的相对表达水平分别为1.02±0.23和1.54±0.51,与癌旁正常组织相比,PSMA3-AS1在NSCLC组织中显著高表达,差异有统计学意义(P<0.05)。对照组、实验1组和实验2组的H1299细胞存活率分别为100%,61%和53%;侵袭数分别为(153.46±13.37),(52.69±5.03)和(42.38±4.28)个;细胞凋亡率分别为(5.38±0.43)%,(12.57±1.04)%和(13.67±0.93)%。对照组与实验1组、实验2组比较,差异均有统计学意义(均P<0.05)。结论PSMA3-AS1在NSCLC组织中高表达,沉默PSMA3-AS1表达,抑制了NSCLC细胞侵袭并促进细胞凋亡。 Objective To investigate the expression of long non-coding RNA(lncRNA) PSMA3-AS1 in non-small cell lung cancer(NSCLC) tissues and its effect on the invasion and apoptosis of NSCLC cells H1299. Methods 50 cases of NSCLC tissue and 50 adjacent normal tissues(as a control group) were selected. Real-time fluorescent quantitative polymerase chain reaction(RT-qPCR) method was used to detect the relative expression level of PSMA3-AS1 in NSCLC tissues and normal tissue adjacent to cancer. H1299 cells were divided into control group, experimental-1 group and experimental-2 group. The control group was transfected with si-NC, experimental-1 group was transfected with si-PSMA3-AS1-1, and experimental-2 group was transfected with si-PSMA3-AS1-2. Cell proliferation was detected by MTT assay. Cell invasion was detected by transwell experiment, and cell apoptosis was detected by flow cytometry. Results The relative expression levels of PSMA3-AS1 in NSCLC tissues and normal tissues adjacent to cancer were 1.02±0.23 and 1.54±0.51,respectively. Compared with normal tissues adjacent to cancer,PSMA3-AS1 was significantly higher in NSCLC tissues,the differences were statistically significant (P<0.05). The survival rates of H1299 cells in the control group,experimental-1 group and experimental-2 group were 100%,61% and 53%,respectively. The invasion numbers of H1299 cells in the control group,experimental-1 group and experimental-2 group were 153.46±13.37,52.69±5.03 and 42.38±4.28,respectively. The apoptosis rates of H1299 cells in the control group,experimental-1 group and experimental-2 group were (5.38±0.43) %, (12.57±1.04) % and (13.67±0.93) %,respectively. The control group was compared with the experimental-1 group or the experimental-2 group respectively,the differences were statistically significant (all P<0.05). Conclusion PSMA3-AS1 is highly expressed in NSCLC tissues. Silencing the expression of PSMA3-AS1 inhibits the invasion of NSCLC cells and promotes cell apoptosis.
