摘要
目的通过对0~6岁难治性肺炎支原体肺炎(RMPP)患儿进行外周血白细胞介素-6(IL-6)及淋巴细胞亚群检测,结合肺炎支原体23SrRNA基因突变位点A2603G或(和)A2604G检测,分析外周血IL-6水平及淋巴细胞亚群与RMPP相关情况。方法肺炎支原体23SrRNA基因突变位点A2603G或(和)A2604G检测采用PCR-Taqman荧光探针技术,IL-6检测方法为流式细胞法。结果研究组肺炎支原体23SrRNA基因突变位点A2603G或(和)A2604G突变率明显高于对照组,差异有统计学意义(P<0.05);研究组各年龄段肺炎支原体23SrRNA基因突变位点A2603G或(和)A2604G突变率比较,差异有统计学意义(P<0.05),年龄越大,其突变比例也越高。研究组各年龄段IL-6水平及其异常情况比较,差异无统计学意义(P>0.05)。结论 RMPP患儿肺炎支原体23SrRNA基因突变位点A2603G或(和)A2604G检测可作为其诊断的常规项目,该检测结果为早期诊断RMPP提供依据。
Objective To investigate the interleukin-6(IL-6) and lymphocyte subsets in peripheral blood of 0-6 year old children with refractory Mycoplasma pneumoniae pneumonia(RMPP),combined with the detection of mutation sites A2603 G or/and A2604 G of 23 SrRNA gene of Mycoplasma pneumoniae, and analyze the levels of IL-6 in peripheral blood and the correlation between lymphocyte subsets and RMPP.Methods Mutation sites A2603 G or/and A2604 G of Mycoplasma pneumoniae 23 SrRNA gene were detected by PCR-Taqman fluorescence probe technique, and IL-6 was detected by flow cytometry.Results The mutation rate of mutation sites A2603 G or/and A2604 G of Mycoplasma pneumoniae 23 SrRNA gene in the study group was significantly higher than that in the control group(P<0.05).The mutation rate of mutation sites A2603 G or/and A2604 G of Mycoplasma pneumoniae 23 SrRNA gene of all age groups in the study group was significantly different(P<0.05),and the older the age, the higher the proportion of mutation.There was no significant difference in IL-6 levels and abnormal conditions among different age groups(P>0.05).Conclusion Detection of 23 SrRNA mutation sites A2603 G or/and A2604 G of Mycoplasma pneumoniae in children with RMPP can be a routine item for diagnosis, and the detection results provide a basis for early diagnosis of RMPP.
