脊髓背角NMDA受体NR2B亚基对舒芬太尼耐受形成的研究 被引量:1

摘要 目的:探讨脊髓背角N-甲基-D-天冬氨酸(NMDA)受体NR2B亚基对舒芬太尼耐受形成的研究。方法:选取40只SD大鼠,根据随机数字表法分为研究组(20例)与对照组(20例),对照组给予皮下注射0.9%氯化钠溶液,研究组给予皮下注射舒芬太尼建立舒芬太尼耐受SD大鼠模型。观察两组SD大鼠注射前及注射后第5、10天的机械性刺激痛阈及热缩足反应潜伏期,并检测注射后第5、10天的脊髓背角NR2B亚基及mRNA表达水平。结果:注射后研究组机械性刺激痛阈值高于对照组(P<0.05);研究组注射后第5天和第10天机械性刺激痛阈值均高于注射前,且注射后第5天高于注射后第10天(P<0.05)。注射后研究组热缩足反应潜伏期值高于对照组(P<0.05);研究组注射后第5天和第10天热缩足反应潜伏期值均高于注射前,且注射后第5天高于注射后第10天(P<0.05)。研究组注射后第5天和第10天脊髓背角NR2B亚基及mRNA水平均高于对照组,且注射后第5天水平均低于注射后第10天(P<0.05)。结论:舒芬太尼耐受形成与脊髓背角NMDA受体NR2B亚基表达水平升高有关。 Objective:To investigate the formation of sufentanil tolerance by the N-methyl-D-aspartate(NMDA)receptor NR2 B subunit of the spinal dorsal horn.Methods:40 SD rats were selected and divided into study group(20 cases)and control group(20 cases)according to the random number table method.The control group took subcutaneous injection of 0.9%sodium chloride solution,and the study group took subcutaneous injection of sufentanil,establishing the sufentanil tolerance SD rat model.The pain threshold of mechanical stimulation and the paw withdrawal thermal latency of two groups of SD rats before injection and on the 5 th and 10 th day after injection were observed,and the NR2 B subunit and the mRNA expression level of the spinal dorsal horn on the 5 th and 10 th day after injection were measured.Results:The pain threshold of mechanical irritation of the study group after injection was higher than that of the control group(P<0.05);the pain threshold of the study group was higher on the 5 th and 10 th day after injection than before the injection,and higher on the 5 th day after injection than the 10 th day after injection(P<0.05).The paw withdrawal thermal latency in the study group after injection was higher than that in the control group(P<0.05);the paw withdrawal thermal latency in the study group was higher on the 5 th and 10 th day after injection than before the injection,and higher on the 5 th day after injection than the 10 th day after injection(P<0.05).The levels of NR2 B subunit and the mRNA of the spinal dorsal horn in the study group were higher than those in the control group on the 5 th and 10 th day after injection,and the levels on the 5 th day after injection were lower than that on the 10 th day after injection(P<0.05).Conclusion:Sufentanil tolerance is related to the increased expression of NMDA receptor NR2 B subunit in the spinal dorsal horn.
