摘要
J亚群禽白血病病毒(ALV-J)是一种严重危害养禽业的肿瘤疾病之一,因目前尚无可供广泛使用的有效疫苗和特效药,防控主要依靠种群净化。本试验以ALV-J原型毒株的gp37基因序列为参照,设计荧光引物,成功建立了特异性较强的ALV-J荧光定量PCR检测方法。随后,将ALV-J病毒液与黄芪提取液先后或同时作用于DF-1细胞,培养72 h后,提取细胞总RNA,用建立的荧光定量PCR方法检测病毒对照组与直接灭活组、预防组、治疗组之间gp37基因表达量的差异,以此探索黄芪对gp37基因转录的影响。结果显示,病毒对照组与直接灭活组、预防组、治疗组的gp37基因表达量均有显著差异(P<0.05),其中预防组的gp37基因表达量与病毒对照组差异最大,表明黄芪能明显下调gp37基因的表达。因此,黄芪可能对J亚型禽白血病的防治有一定作用。
Avian leukosis virus subgroup J(ALV-J)is a serious cancer-causing virus in poultry,for which no effective vaccines and specific drugs are currently available,and prevention and control rely mainly on population purification.In this study,fluorescent primers were designed from the gp37 gene sequence of the prototype ALV-J strain and used in a fluorescent quantitative PCR for specific detection of ALV-J.ALV-J solution and Astragalus extract were added separately or successively to DF-1 cells.After 72 h incubation,total RNA was extracted from DF-1 cells of the virus control group,direct inactivation group,prevention group,and treatment group,and detected by the established fluorescence quantitative PCR.The difference of gp37 gene expression among the groups was used to explore the influence of Astragalus on the transcription of gp37 gene.The results showed that there were significant differences in the expression of gp37 gene among the virus control group and the direct inactivation group,the prevention group,the treatment group(P<0.05),and that the gp37 gene expression in the prevention group differed significantly from the virus control group,indicating that Astragalus can significantly down-regulate the expression of gp37 gene.Thus,Astragalus may have potential for prevention and treatment of J subtype avian leukemia.
