摘要
在人肺癌H460及H460/DDP细胞中,通过干扰长链非编码RNA SLC7A11-AS1来探究其对细胞耐药的影响.通过大数据库分析SLC7A11-AS1与肺癌患者生存期的关系,利用实时定量PCR检测SLC7A11-AS1在人肺癌细胞H460和顺铂耐药细胞株H460/DDP中的表达.在H460/DDP细胞中瞬时转染SLC7A11-AS1的干扰片段,MTT法检测H460/DDP对顺铂药物敏感性的变化.结果表明,SLC7A11-AS1表达量高的肺癌患者生存期明显短于SLC7A11-AS1表达量低的肺癌患者(P<0.05);SLC7A11-AS1的表达在H460/DDP细胞中明显上调(P<0.05);干扰SLC7A11-AS1表达后,H460/DDP细胞对顺铂的化疗敏感性明显提高.因此,SLC7A11-AS1与肺癌的不良预后相关并影响肺癌的化疗耐药.
Interference of long non-coding RNA SLC7 A11-AS1 in human lung cancer H460 and H460/DDP cells was used to investigate the effect of SLC7 A11-AS1 on cell drug resistance.The relationship between SLC7 A11-AS1 and survival of lung cancer patients was analyzed by large database.The expression of SLC7 A11-AS1 in human lung cancer cell line H460 and cisplatin-resistant cell line H460/DDP was detected by real-time quantitative PCR.The interference fragment of SLC7 A11-AS1 was transient transfected into H460/DDP cells.The sensitivity of H460/DDP to cisplatin was detected by MTT assay.The results showed that the survival of lung cancer patients with high SLC7 A11-AS1 expression was significantly shorter than that of lung cancer patients with low SLC7 A11-AS1 expression(P<0.05).The expression of SLC7 A11-AS1 was significantly up-regulated in H460/DDP cells(P<0.05).After the expression of SLC7 A11-AS1 was interfered,the sensitivity of H460/DDP cells to cisplatin wassignificantly increased.Therefore,SLC7 A11-AS1 is associated with poor prognosis of lung cancer and affects chemotherapy resistance of lung cancer.
